摘  要:目的观察电针结合美多巴对帕金森病(PD)模型大鼠黑质酪氨酸羟化酶(TH)和胶质细胞源性神经营养因子(GDNF)表达的影响,探究针药结合在帕金森病治疗中的可能作用机制。方法于2019年6月开展实验研究,选择30只SD大鼠随机分为正常组、模型组、药物组、电针组和针药结合组,每组各6只。采用单侧纹状体微量注射6-羟基多巴胺(6-OHDA)制备PD模型,药物组美多巴悬浊液灌胃;电针组高频电针(100 Hz)治疗;针药结合组先美多巴悬浊液灌胃,后高频电针治疗;三组均治疗2个疗程(每天1次,7天为1个疗程)。免疫组织化学法检测黑质TH、GDNF的表达。结果 PD大鼠黑质TH表达减少,美多巴、电针和针药结合治疗都可增加黑质TH和GDNF表达(P<0.01),但针药结合治疗效果优于单用美多巴或电针(P<0.05)。结论针药结合治疗PD大鼠可增加其黑质TH、GDNF的表达,效果优于单用美多巴或电针。Objective To observe the effect of electroacupuncture combined with madopar therapy on the expression of tyrosine hydroxylase and glial cell line-derived neurotrophic factor in Parkinson’s disease(PD) rats, and to explore the possible mechanism of electroacupuncture combined with drug therapy in the treatment of PD.Methods The experimental study was conducted in June 2019, thirty SD rats were randomly divided into five groups: control group, model group, drug group, electroacupuncture group and electroacupuncture combined with drug group, 6 rats in each group. PD model was conducted by micro-injection of 6-hydroxydopamine(6-OHDA) into unilateral striatum, and the rats in the drug group were given madopar suspension through intragastric administration, the rats in the electroacupuncture group were given high frequency electroacupuncture(100 Hz), the rats in the electroacupuncture combined with drug group were given madopar suspensionintragastric administration followed by electroacupuncture for treatment. The three treatment groups were treated for 2 courses(1 time/day,7 days/course). Immuno-histochemistry was used to detect the expression of TH and GDNF in substantia Nigra.Results The expressions of TH and GDNF in substantia nigra of PD rats were decreased, however,the expressions of TH and GDNF in substantia Nigra of PD rats were increased after the treatment of Madopar,electroacupuncture and electroacupuncture combined with drug(P<0.01), and electroacupuncture combined with drug was superior to Madopar or electroacupuncture alone(P<0.05).Conclusions The electroacupuncture combined with drug could increase the expression of TH and GDNF in Substantia Nigra,and the effect was better than that of madopar or electroacupuncture alone.
