摘 要:目的研究阿里红多糖(Fomes officinalis polysaccharides,FOPS)对阿尔茨海默病(Alzheimer’s disease,AD)模型大鼠海马区神经元的形态改变及脑皮层和海马区氧化应激水平的影响。方法72只健康雄性SD大鼠称重并随机分为对照组、模型组、盐酸多奈哌齐组(0.5mg/kg)、阿里红多糖高、中、低剂量组(100、50、25 mg/kg),每组12只。采用大鼠双侧海马CA1区注射(5μL/侧)Aβ1-42建立AD大鼠模型,给药30 d后,苏木精-伊红(hematoxylin-eosin staining,HE)染色观察海马神经元的形态改变,DCFH-DA荧光探针分析处理对各组大鼠海马区和脑皮质层中活性氧簇(reactiveoxygenspecies,ROS)的影响,酶联免疫吸附(enzymelinked immunosorbentassay,ELISA)法检测8羟基脱氧鸟苷(8-hydroxy-2’-deoxyguanosine,8-OHdG)、3-硝基酪氨酸(3-nitrotyrosine,3-NT)、4羟基壬烯醛(4-hydroxy-2-nonenal,4-HNE)的含量。结果实验30 d后,与模型组比较,盐酸多奈哌齐及阿里红多糖组大鼠锥体细胞数目明显增多、形态较完整、海马神经元大多数较正常、排列较规整紧密、着色均匀、核仁清晰,大鼠海马区及脑皮层ROS含量明显降低(P<0.01),8-OHdG、3-NT、4-HNE含量明显降低(P<0.01)。结论阿里红多糖通过影响Aβ1-42诱导的AD大鼠脑内ROS、8-OHdG、3-NT、4-HNE含量,改善脑内氧化应激状态,起到改善认知障碍和保护神经的作用。Objective To research the effect of Fomes officinalis polysaccharides on the morphological changes of hippocampal neurons in Alzheimer’s disease(AD)model rats and the level of oxidative stress in the cerebral cortex and hippocampus.Methods Total of 72 eligible male SD rats were weighed and randomly divided into control group,AD model group,donepezil hydrochloride group(0.5 mg/kg),FOPS high,medium and low dose groups(25,50,100 mg/kg),with 12 rats in each group.The AD rat model was established by injecting(5μL/side)Aβ1-42 in the hippocampal CA1 area of both sides of the rat.After 30 days of treatment,hematoxylin-eosin staining(HE)staining was used to observe the morphological changes of hippocampal neurons.2,7-dichlorodi-hydrofluorescein diacetate fluorescent probe was used to analyze reactive oxygen species and ELISA method was used to detect the content of 8-OHdG,3-NTand 4-HNEin the hippocampus and cerebral cortex of rats in each group.Results After 30 days,compared with the model group,the number of pyramidal cells in donepezil hydrochloride group and Fomes officinalis polysaccharides group was significantly increased,the morphology was more complete,the majority of hippocampal neurons were more normal,the arrangement was more regular and compact,the color was uniform and the nucleolus was clear.The levels of ROS hippocampus and cerebral cortex of rats were significantly reduced(P<0.01),and the levels of 8-OHdG,3-NT,and 4-HNE were significantly reduced(P<0.01).Conclusion FOPS improves the oxidative stress state in the brain by affecting the content of ROS,8-OHdG,3-NT,4-HNE of AD rats induce by Aβ1-42,and plays a role in improving cognitive impairment and protecting nerves.
