摘  要:为探究玉米赤霉烯酮(ZEA)及脱氧雪腐镰刀菌烯醇(DON)对动物CD4^+T细胞活化的影响,以李斯特菌感染小鼠的CD4^+T细胞为研究对象,研究了低剂量的ZEA、DON对小鼠T细胞活化标志分子CD69、CD71以及CD44、CD223的影响。试验分别设空白组(Control)、李斯特菌组(Listeria)、ZEA+李斯特菌组(ZEA组)、DON+李斯特菌组(DON组)、ZEA+DON+李斯特菌组(联合毒素组),用流式细胞术分别检测李斯特菌感染第3天的小鼠的脾CD4^+T细胞CD69的表达及感染第7天小鼠的脾CD4^+T细胞CD71、CD44、CD223的表达。结果显示,与空白组相比,李斯特菌组CD4^+T细胞CD69、CD71及CD44、CD223表达均显著或极显著升高(P<0.05或P<0.001)。与李斯特菌组相比,ZEA组CD69、CD71和CD44表达均无显著差异(P>0.05),CD223的表达显著下降(P<0.05);DON组CD71和CD223的表达均无显著差异(P>0.05),CD69、CD44表达均极显著下降(P<0.001);联合毒素组CD69、CD71和CD223表达均无显著差异(P>0.05),CD44表达极显著下降(P<0.01)。与ZEA组相比,联合毒素组CD69、CD71和CD44、CD223表达均无显著差异(P>0.05)。与DON组相比,联合毒素组CD71和CD223表达均无显著差异(P>0.05),CD44和CD69的表达显著或极显著升高(P<0.05或P<0.001)。结果表明,低剂量的ZEA、DON及其联合均能不同程度地抑制李斯特菌感染小鼠的CD4^+T细胞活化标志分子的表达,进而抑制细胞的活化,对动物具有免疫抑制作用;低剂量的ZEA和DON联合应用在对小鼠CD4^+T细胞活化影响上具有一定的拮抗作用。In order to explore the effects of zearalenone and deoxynivalenol on the activation of animal CD4^+T cells.In this experiment,CD4^+T cells from listeria-infected mouse were used to study the effects of low-dose ZEA and DON on mouse T cell activation marker molecules CD69,CD71,CD44 and CD223.The control group,Listeria group,ZEA+Listeria group,DON+Listeria group and ZEA+DON+Listeria group were set respectively.Flow cytometry was used to detect the expression of CD4^+T cells CD69 in the spleen on day 3 and CD71,CD44 and CD223 on day 7 after listeria infection.The results showed that,compared with the control group,the expression of CD69,CD71,CD44 and CD223 of CD4^+T cells in Listeria group was significantly or extremely significantly increased(P<0.05 or P<0.001).Compared with Listeria group,the expression of CD69,CD71 and CD44 in ZEA group had no significant difference(P>0.05).The expression of CD223 decreased significantly(P<0.05).The expression of CD71 and CD223 in DON group had no significant difference(P>0.05),while the expressions of CD69 and CD44 decreased extremely significantly(P<0.001).The expression of CD69,CD71 and CD223 in the combined group had no significant difference(P>0.05),while the expression of CD44 decreased extremely significantly(P<0.01).Compared with ZEA group,the expressions of CD69,CD71,CD44,and CD223 in the combined group had no significant difference(P>0.05).Compared with DON group,the expressions of CD71 and CD223 in the combined group had no significant difference(P>0.05),while the expressions of CD44 and CD69 increased significantly or extremely significantly(P<0.05 or P<0.001).The results showed that low-dose ZEA,DON and their combination could inhibit the expression of CD4^+T cell activation marker molecules in listeria-infected mouse to varying degrees.It also inhibit the activation of cells,which had immunosuppressive effect on animals.Besides,the low-dose ZEA and DON have a certain antagonistic effect on mouse CD4^+T cell activation.
