摘 要:目的探讨血管内皮生长因子(VEGF)基因的遗传多态性与肾细胞癌(RCC)的关系。方法搜索Web of Science、Pub Med和Embase数据库的相关文章,共2309名RCC患者和3337名对照的7个病例对照研究,分析VEGF基因+936C>T和+405C>G多态性的定性数据。结果VEGF基因+936C>T多态性与RCC风险相关(加成模型:OR=1.38,95%CI=1.11~1.72,P=0.004;显性模型:OR=1.18,95%CI=1.03~1.36,P=0.02;隐性模型:OR=1.28,95%CI=1.04~1.57,P=0.02;等位基因模型:OR=1.20,95%CI=1.07~1.34,P=0.001)。然而,比较4种遗传模型,VEGF+405C>G多态性与RCC无相关性(加成模型:OR=0.77,95%CI=0.48~1.23,P=0.27;优势模型:OR=0.93,95%CI=0.73~1.18,P=0.54;优势模型:OR=0.84,95%CI=0.67~1.05,P=0.12;等位基因模型:OR=1.02,95%CI=0.79~1.32,P=0.87)。结论VEGF+936C>T多态性与RCC风险相关,而与VEGF+405C>G多态性无关。Objective To identify the relationships between the genetic polymorphisms in the vascular endothelial growth factor(VEGF)gene and the risk of renal cell carcinoma.Methods The Web of Science,Pub Med and Embase databases were searched for relative articles.7 case–control studies with a total of 2309 patients with renal cell carcinoma(RCC)and 3337 control subjects were included to qualitative data analysis for VEGF gene+936C>T and+405C>G polymorphism.Results The pooled results showing evidence of association between the VEGF gene+936C>T polymorphism and RCC risk(additive model:OR=1.38,95%CI=1.11–1.72,P=0.004;dominant model:OR=1.18,95%CI=1.03–1.36,P=0.02;recessive model:OR=1.28,95%CI=1.04–1.57,P=0.02;allelic model:OR=1.20,95%CI=1.07–1.34,P=0.001).However,there was no significant association between VEGF+405C>G polymorphism and RCC comparing 4 genetic model(additive model:OR=0.77,95%CI=0.48–1.23,P=0.27;codominant model:OR=0.93,95%CI=0.73–1.18,P=0.54;dominant model:OR=0.84,95%CI=0.67–1.05,P=0.12;allelic model:OR=1.02,95%CI=0.79–1.32,P=0.87).Conclusion The results indicated that VEGF+936C>T polymorphism,but not VEGF+405C>G polymorphism was associated with the risk of renal cell carcinoma.
