摘 要:目的探讨卡瓦胡椒素B对人舌鳞癌移植瘤生长的影响。方法构建人舌鳞癌移植瘤裸鼠模型,随机分为4组:对照组、观察组(低、中、高剂量组)。观察并测量移植瘤的体积和质量;TUNEL检测移植瘤的凋亡情况;Western blotting检测凋亡相关蛋白和血管内皮生长因子(VEGF)的表达;免疫组织化学染色法检测CD34的表达。结果28 d后,观察组裸鼠移植瘤的体积和质量均显著低于对照组,且高剂量组明显低于中、低剂量组(P<0.05)。观察组TUNEL染色阳性强度明显高于对照组。观察组Bax、Cleaved Caspase-3水平显著高于对照组,Bcl-2水平显著低于对照组(P<0.05);且高剂量组Bax、Cleaved Caspase-3水平显著高于中、低剂量组,Bcl-2水平显著低于中、低剂量组(P<0.05)。观察组CD34染色阳性强度明显低于对照组。观察组VEGF水平显著低于对照组,且高剂量组显著低于中、低剂量组(P<0.05)。结论卡瓦胡椒素B可抑制人舌鳞癌移植瘤的生长,促进肿瘤细胞凋亡,抑制肿瘤血管生成。Objective To explore the effects of Flavokawain B on the xenograft tumor growth of human tongue squamous cell carcinoma.Methods A nude mouse model of human tongue squamous cell carcinoma was constructed and randomly divided into four groups:control group and observation groups(low-dose group,middle-dose group,high-dose group).The volume and weight of the transplanted tumor were measured.The apoptosis of the transplanted tumor was detected by TUNEL method.The expressions of apoptosis-related proteins and vascular endothelial growth factor(VEGF)were detected by Western blotting.The expression of CD34 was detected by immunohistochemical staining.Results After 28 days,the volume and weight of the transplanted tumor in the observation groups were lower than in the control group,and were also lower in the high-dose group than in the middle-and low-dose groups(P<0.05).The positive intensity of TUNEL staining in observation groups was significantly higher than in control group.The expressions of Bax and cleaved caspase-3 were higher in observation groups than in control group,but the expression of Bcl-2 was lower in observation groups than in control group(P<0.05).The expressions of Bax and cleaved caspase-3 in the high-dose group were higher than in the middle-and low-dose groups,and the expression of Bcl-2 was lower than in the middle-and low-dose groups(P<0.05).The positive intensity of CD34 staining in observation groups was significantly lower than in control group.The expression of VEGF in the observation groups was lower than in the control group,and it was lower in the high-dose group than in the middle-and low-dose groups(P<0.05).Conclusion Flavokawain B can inhibit the xenograft tumor growth of human tongue squamous cell carcinoma,promote its apoptosis and inhibit its angiogenesis.
