摘  要:目的探讨紫铆因(Butein)介导mTOR信号通路对宫颈癌HeLa细胞侵袭、迁移的影响。方法将HeLa细胞分为对照组(无药物干预)和观察组(10、20、40μmol·L^-1 Butein干预),分别采用MTT、Transwell、划痕实验、免疫印迹法及PCR检测不同浓度Butein对HeLa细胞活力、侵袭、迁移和mTOR信号通路的影响。结果对照组细胞存活率为100%,与不同浓度观察组作用不同时间(24、36、48、72 h)的细胞存活率比较,差异有统计学意义(P<0.05),且细胞存活率与Butein浓度及干预时间呈负相关。观察组细胞侵袭数和迁移数均显著低于对照组(P<0.05),其中,40μmol·L^-1组最低,10μmol·L^-1组最高,组间差异均有统计学意义(P<0.05)。观察组mTOR/P70S6K mRNA和蛋白表达水平均显著低于对照组(P<0.05),其中,40μmol·L^-1组最低,10μmol·L^-1组最高,组间差异均有统计学意义(P<0.05)。结论Butein可抑制宫颈癌HeLa细胞增殖、迁移和侵袭,并呈时间和浓度依赖性,其作用机制可能与抑制mTOR/P70S6K信号通路活化有关。Objective To investigate the effects of butein-mediated mTOR signal pathway on the invasion and migration of HeLa cells.Methods HeLa cells were divided into control group(no drug intervention)and observation groups(10,20,40μmol·L^-1 butein intervention).MTT,transwell,scratch,immunoblotting and PCR were applied to detect the effects of different concentrations of butein on the viability,invasion,migration of HeLa cells and mTOR signal pathway.Results The cell survival rate was 100%in the control group.There were significant differences in the cell survival rate between the control group and observation subgroups of different butein concentrations at different time points(24,36,48 and 72 h)(P<0.05).Moreover,the survival rate of HeLa cells was negatively correlated with the concentration and treating time of butein.In addition,observation groups had less invasion and migration cells than the control group(P<0.05).Among the observation groups,the subgroup of 40μmol·L^-1 butein had the least invasion and migration cells,and the 10μmol·L^-1 butein subgroup had the most invasion and migration cells(P<0.05).The mRNA and protein expression levels of mTOR/P70S6K in the observation groups were all lower than in the control group(P<0.05),and they were the lowest in the subgroup of 40μmol·L^-1 butein and highest in the subgroup of 10μmol·L^-1 butein(P<0.05).Conclusion Butein can inhibit the proliferation,migration and invasion of HeLa cells in a time-and concentration-dependent way,and the mechanism may be related to the inhibition of mTOR signal activation.
