摘 要:目的佛手药材中香叶木苷和橙皮苷含量测定方法的建立。方法2015年9月至2018年6月,以DiamonsilC18(250mm×4.6 mm,5μm)为色谱柱,乙腈(A)-0.1%醋酸水(B)(20∶80)等度洗脱为流动相,285 nm为检测波长,1.0 m L/min的流速,25℃的柱温,10μL的进样量。结果香叶木苷、橙皮苷进样量分别在12.32~616 ng(r=0.9998),8.128~1016 ng(r=0.9999)范围内与峰面积呈良好线性关系,平均回收率(n=6)分别为99.32%、100.87%;相对标准偏差(RSD)分别为2.29%、2.24%。结论该法简便、准确、可靠,可作为佛手药材中香叶木苷、橙皮苷含量测定的有效方法。Objective Establishment of a method for determination of diosmin and hesperidin in bergamot medicinal materials.Methods From September 2015 to June 2018,the chromatographic column was Diamonsil C18(250 mm×4.6 mm,5μm),mobile phase was acetonitrile(A)-0.1%acetic acid water(B)inisometric elution(20:80).The detection wavelength was 285 nm and the flow rate wass 1.0 mL min-1 at 25℃,and the injection volume was 10μL.Results The contents of diosmin and hesperidin had good linear relationship with the peak area in the ranges of 12.32~616 ng(r=0.9998)and 8.128~1016 ng(r=0.9999)respectively,and the average recoverie rate(n=6)of five components was 99.32%with RSD of 2.29%and 100.87%with RSD of 2.24%respectively.Conclusion The method is simple,accurate and reliable,and can be used as an effective method for the determination of diosmin and hesperidin in bergamot.
