摘 要:目的:探讨有氧运动对APP/PS1小鼠大脑皮质和海马组织线粒体氧化磷酸化功能的影响,为有氧运动防治阿尔茨海默症提供理论依据。方法:雄性APP/PS1小鼠随机分为安静对照组(T-CG组)和运动组(T-EG组),野生型C57BL/6J小鼠也随机分为安静对照组(W-CG组)和运动组(W-EG组),T-EG和W-EG组小鼠进行为期8周的跑台训练。在第9周进行Morris水迷宫实验检测所有小鼠的认知能力。水迷宫实验结束后,分离所有小鼠大脑皮质和海马组织。通过试剂盒检测大脑皮质和海马组织ATP含量、线粒体呼吸链复合体Ⅰ~Ⅴ相对酶活性和线粒体膜电位情况;通过Western blot检测线粒体Cox5a、Cox2、Nd1和Sdhc的蛋白水平。结果:与W-CG组相比,T-CG组小鼠大脑皮质和海马组织ATP含量、线粒体呼吸链复合体Ⅰ~Ⅴ相对酶活性、线粒体膜电位以及Cox5a、Cox2、Nd1和Sdhc的蛋白水平均显著下降;Morris水迷宫实验逃逸时间和逃逸距离显著增加。而经过8周跑台训练后,与T-CG组相比,T-EG组小鼠大脑皮质和海马组织ATP含量显著上升;大脑皮质线粒体呼吸链复合体Ⅰ、Ⅲ、Ⅳ和Ⅴ的相对酶活性显著提高,而海马组织线粒体呼吸链复合体Ⅰ~Ⅴ的相对酶活性均显著提高;大脑皮质和海马组织线粒体膜电位显著上升;线粒体Cox5a、Cox2、Nd1和Sdhc的蛋白水平均显著上升;Morris水迷宫实验逃逸时间和逃逸距离显著减少。此外,与W-CG组相比,W-EG组小鼠大脑皮质和海马组织ATP含量、线粒体呼吸链复合体Ⅰ-Ⅴ相对酶活性、线粒体膜电位以及Cox5a、Cox2、Nd1和Sdhc的蛋白水平均显著提高;Morris水迷宫实验逃逸时间显著减少。结论:有氧运动能够显著提高APP/PS1小鼠大脑皮质和海马组织ATP含量、线粒体呼吸链复合体酶活性和线粒体膜电位,通过提高线粒体氧化磷酸化调控关键蛋白表达,改善APP/PS1小鼠大脑皮质和海马组织线粒体氧化磷酸化功能,提高认知功能�Objective To investigate the effect of aerobic exercise on the oxidative phosphorylation of mitochondria in the cortex and hippocampus of APP/PS1 mice,so as to provide theoretical basis for preventing Alzheimer’s disease through aerobic exercise.Methods APP/PS1 male mice and wild-type C57 BL/6 J male ones were randomly divided into a control group(T-CG and W-CG)and an exercise group(T-EG and W-EG)respectively.The mice in the exercise groups underwent an 8-week treadmill training.All mice were conducted Morris water maze experiment at the 9th week to test the cognitive function,and then their cerebral cortex and hippocampus were isolated.The content of adenosine triphosphate(ATP),the relative enzyme activity of mitochondrial respiratory chain complexⅠ~Ⅳand the mitochondrial membrane potential were detected using the kit.Moreover,the protein levels of cytochrome c oxidase subunit 5 a(Cox5 a),cytochrome c oxidase subunit 2(Cox2),NADH dehydrogenase subunit 1(Nd1)and succinate dehydrogenase complex subunit C(Sdhc)were assayed using Western blotting.Results Compared with the W-CG group,in the T-CG group the content of ATP,the relative enzyme activity of mitochondrial respiratory chain complexesⅠ~Ⅴ,the mitochondrial membrane potential and the protein levels of Cox5 a,Cox2,Nd1 and Sdhc in the cerebral cortex and hippocampus decreased significantly,while the average escape time and distance increased significantly in the Morris water maze experiment.After 8-week treadmill exercise,compared with the T-CG group,in the T-EG group all measurements increased significantly except that the average escape time and disdance decreased significantly.Conclusion Aerobic exercise can significantly improve the content of ATP,the enzyme activity of mitochondrial respiratory chain complex and the mitochondrial membrane potential in the cortex and hippocampus of APP/PS1 mice.Moreover,aerobic exercise can significantly enhance their mitochondrial oxidative phosphorylation levels and improve their cognitive function by increasing t
