摘  要:目的:本研究拟利用网络药理学方法探讨青黄散治疗骨髓增生异常综合症(MDS)的潜在活性成分和作用机制。方法:通过BATMAN-TCM平台收集雄黄、青黛的活性成分和作用靶点信息,GeneCards和PALM-IST平台收集MDS疾病靶点基因;采用Cytoscape 3.2.1构建“药物-活性成分-靶点”网络并利用network analyzer插件进行拓扑结构分析,采用STRING数据库进行基因互作分析,采用Metascape数据库进行基因本体论(GO)分析及KEGG信号通路富集分析。结果:共获得青黄散治疗MDS的潜在活性成分4个,其中自由度≥2的重要靶基因54个;基因富集分析显示青黄散主要通过调控氧化应激、炎症反应、能量代谢和细胞凋亡等生物学过程发挥治疗MDS的作用。结论:本研究从网络药理学角度初步阐释了青黄散治疗MDS的潜在活性成分和作用机制,揭示了其多成分、多靶点、多途径调节的特点,为后续机制研究的开展提供了基础和依据。Objective:To explore the potential active components and mechanism of Qinghuang Powder on the treatment of myelodysplastic syndrome(MDS)through network pharmacology.Methods:The BATMAN-TCM platform was used to collect the active components and target information of Realgar and Indigo Naturals.The gene of MDS was collected by GeneCards and PALM-IST platform.The"drug-active components-target"network was constructed and visualized by Cytoscape 3.2.1.The network topology was analyzed using the network analyzer.The STRING database was used for gene interaction analysis.The Metascape database was used for gene ontology(GO)analysis and KEGG signal pathway enrichment analysis.Results:As a result,54 important target genes with degrees of freedom≥2.Gene enrichment analysis showed that Qinghuang Powder plays a role in the treatment of MDS by regulating Oxidative stress,inflammatory response,energy metabolism and apoptosis.Conclusion:The study preliminarily explained the potential active components and mechanism of Qinghuang Powder on the treatment of MDS based on network pharmacology,revealed the effect characteristics of which was multi-component,multi-target and multi-channel integration regulation,and provided the basis for further experimental research.
