卵巢癌患者腹水中MDSCs通过靶向VASP促进卵巢癌干细胞表型及细胞增殖

摘　　要：目的探究卵巢癌腹水中髓系来源抑制细胞(myeloid-derived suppressor cells,MDSCs)对SKOV 3细胞的影响及机制。方法取20例卵巢癌患者的腹水,使用磁珠分选的技术分离腹水中MDSCs细胞,并用流式细胞术鉴定CD 33和CD 11 b的表达;将SKOV 3细胞分别与磷酸缓冲盐溶液(phosphate-buffered saline,PBS)和MDSCs上清共孵育,qPCR和Western blot检测孵育后SKOV 3细胞干性指标的表达,以及血管扩张刺激磷蛋白(vasodilator stimulated phosphoprotein,VASP)的表达,CCK 8检测细胞增殖能力的变化;体外抑制SKOV 3细胞VASP表达的同时与MDSCs细胞上清孵育,检测细胞干性的变化以及细胞增殖能力的变化。结果采用磁珠分选的MDSCs表达CD 33和CD 11 b,符合MDSCs的特征;将SKOV 3细胞与MDSCs上清共孵育后细胞干性指标CD 44、NANOG、OCT-4表达上调(P<0.001),VASP表达亦上调(P<0.001),细胞增殖显著增加(P<0.05);抑制VASP表达后,MDSCs上清对SKOV 3细胞干性及增殖能力的影响被逆转。结论卵巢癌腹水中MDSCs能够调控VASP的表达而影响SKOV 3细胞干性和增殖。Objective To investigate the effect and mechanism of MDSCs on ascites in ovarian cancer on SKOV 3 cells.Methods Ascites from 20 ovarian cancer patients were collected,and MDSCs were isolated using magnetic bead sorting technology,and the expression of CD 33 and CD 11 b were identified by flow cytometry.SKOV 3 cells were co-incubated with PBS and MDSCs supernatants,and qPCR was performed.Western blot was used to detect the expression of SKOV 3 cell dryness index and VASP expression,and CCK 8 was used to detect the change of cell proliferation ability.In vitro inhibition of SKOV 3 cell VASP expression was incubated with the supernatant of MDSCs to detect changes in cell dryness and cells Changes in proliferative capacity.Results The magnetic beads sorted MDSCs expressed CD 33 and CD 11 b,which accorded with the characteristics of MDSCs.After co-incubation of SKOV 3 cells with the supernatant of MDSCs,the expression of CD 44,NANOG and OCT-4 were up-regulated(P<0.001),and VASP was expressed.It was also up-regulated(P<0.001),and the cell proliferation was significantly increased(P<0.05).After inhibiting the expression of VASP,the effect of MDSCs supernatant on the stemness and proliferation ability of SKOV 3 cells was reversed.Conclusion MDSCs in ovarian cancer ascites can regulate the expression of VASP and affect the stemness and proliferation of SKOV 3 cells.
