摘  要:目的研究白藜芦醇(resveratrol,Res)通过下调核因子κB(nuclear factor kappa-B,NF-κB)P65对人肺腺癌A549细胞增殖的影响。方法使用细胞计数试剂盒-8(CCK-8)测定A549细胞的增殖;选用逆转录-聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)检测Res对A549细胞NF-κB P65 mRNA表达的影响;采用流式细胞仪分析A549细胞的细胞周期及凋亡;用蛋白免疫印迹法(Western blotting)检测A549细胞NF-κB P65蛋白的表达。结果 Res可通过下调NF-κB P65抑制人肺腺癌A549细胞的增殖,阻止A549细胞周期、促进细胞凋亡,同时抑制其蛋白水平的表达。结论 Res通过下调NF-κB P65可以明显抑制人肺腺癌A549细胞的增殖。Objective Resveratrol(Res) was used to down-regulate the proliferation of human lung adenocarcinoma A549 cells by down-regulating nuclear factor kappa B(NF-κB)P65.Methods The proliferation of A549 cells was determined by using Cell Counting Kit-8(CCK-8).Reverse Transcription-Polymerase Chain Reaction(RT-PCR) was used to detect the effect of Res on the expression of NF-κB P65 mRNA in A549 cells.The cell cycle and apoptosis of A549 cells were analyzed by flow cytometry.The NF-κB P65 protein expression in A549 cells was detected by Western blotting.Results Res can inhibit the proliferation of human lung adenocarcinoma A549 cells by down-regulating NF-κB P65,prevent A549 cell cycle,promote cell apoptosis and inhibit the expression of protein.Conclusion Res can significantly inhibit the proliferation of human lung adenocarcinoma A549 cells by down-regulating NF-κB P65.
