美沙拉嗪治疗溃疡性结肠炎的有效浓度分析研究

摘　　要：目的:研究美沙拉嗪肠溶片在不同严重程度溃疡性结肠炎患者体内药物的起始有效浓度及明显增加不良反应风险的最低浓度。方法:2017年7月-2019年6月收治活动期溃疡性结肠炎患者95例,根据改良Mayo评分法分型分为轻、中、重三组,均予美沙拉嗪口服治疗。分别于入组后第1周、2周、1个月、3个月、6个月、1年测定血浆中美沙拉嗪浓度,并评估患者缓解情况和不良反应。结果:轻度患者治疗2周起效,最低有效浓度为(2.556±0.627)mg/L;中度患者2周起效,最低有效浓度为(2.838±0.518)mg/L;重度患者1个月起效,最低有效浓度为(3.199±0.497)mg/L。三组不良反应发生率比较,差异无统计学意义(P>0.05)。结论:美沙拉嗪治疗溃疡性结肠炎具有良好的有效性和安全性。Objective:To investigate the initial effective concentration of mesalazine enteric coated tablets in patients with different severity of ulcerative colitis and the lowest concentration that significantly increases the risk of adverse reactions.Methods:From July 2017 to June 2019,95 patients with active ulcerative colitis were selected.According to the modified Mayo score,they were divided into three groups:mild,moderate and severe.They were treated with oral mesalazine.The concentration of 5-ASA in plasma was measured at 1 week,2 weeks,1 month,3 months,6 months and 1 year after admission,the remission and adverse reactions were evaluated.Results:Mild patients took effect after 2 weeks of treatment,the lowest effective concentration was(2.556±0.627)mg/L.Moderate patients took effect after 2 weeks of treatment,the lowest effective concentration was(2.838±0.518)mg/L.Severe patients took effect after 1 month of treatment,the lowest effective concentration was(3.199±0.497)mg/L.There was no significant difference in the incidence of adverse reactions among the three groups(P>0.05).Conclusion:Mesalazine has good efficacy and safety in the treatment of ulcerative colitis.
