摘  要:目的检测胍丁胺酶(Agmatinase,AGMAT)在肺腺癌组织中的表达及其对肺腺癌细胞增殖功能的影响。方法运用Oncomine数据库分析肺腺癌中AGMAT的mRNA表达,采用Western blot方法检测肺腺癌细胞系中AGMAT的蛋白表达。通过Kaplan-Meier Plotter数据库分析AGMAT基因与肺癌患者预后的关系。应用化学合成小干扰RNA技术沉默肺腺癌细胞中AGMAT的表达,并采用Western blot实验验证基因沉默效率。采用CCK-8和克隆形成实验检测转染siAGMAT后对肺腺癌细胞增殖能力的影响。结果肺癌组织中AGMAT mRNA表达水平显著上调,肺腺癌细胞系A549和H1299中AGMAT蛋白表达水平显著高于正常支气管上皮细胞HBE,且AGMAT的高表达与肺癌患者的临床不良预后显著相关;siRNA干扰AGMAT表达能够显著抑制肺腺癌细胞A549和H1299的增殖能力(P<0.01)。结论AGMAT在肺腺癌组织和细胞中高表达,是潜在的治疗靶点。Objective To study the expression of Agmatinase(AGMAT)in lung adenocarcinoma and its effect on the proliferation function of lung adenocarcinoma cells.Methods Oncomine database was used to analyze the expression of AGMATmRNA,and Western blot was used to detect the expression of AGMAT protein in lung adenocarcinoma cell lines.Kaplan-Meier Plotter database was used to analyze the relationship between AGMAT gene and prognosis of patients with lung cancer patients.The chemical synthesis of small interfering RNA technology was used to silence the expression of AGMAT gene,and Western blot used to verify the gene silencing efficiency.CCK-8 and colony formation were used to detect the effect of siAGMAT transfection on the proliferation of lung adenocarcinoma cells.Results The expression level of AGMAT mRNA in lung cancer tissue was significantly upregulated,and also higher in lung adenocarcinoma A549 and H1299 cells than in normal bronchial epithelial cells for AGMAT protein,and the high expression of AGMAT was related positively to the clinical prognosis of patients with lung cancer(P<0.01).siRNA interference significantly inhibited the proliferation of lung adenocarcinoma A549 and H1299 cells(P<0.01).Conclusion AGMAT is involved in the pathogenesis of lung adenocarcinoma,a potential therapeutic target.
