摘　　要：目的探讨泛素连接酶Cbl-b是否通过调节c-Met影响肺癌放射敏感性。方法采用0Gy,4Gy,8Gy以及12Gy的放射线作用肺癌细胞A549,采用CCK-8法检测其吸光度值,计算细胞活力;采用细胞克隆形成实验检测放射敏感性;采用Western blot法检测泛素连接酶Cbl-b蛋白表达水平。将空白对照质粒(Control组)和Cbl-b过表达质粒(Cbl-b组)转染进细胞,采用CCK-8和细胞克隆形成实验检测放射敏感性;采用Western blot法检测c-Met蛋白表达水平。结果4Gy,8Gy以及12Gy的放射线显著抑制细胞增殖,并且随着剂量增加细胞增殖能力显著下降。放射线同样能抑制A549细胞的克隆形成率,并且随着放射线剂量增加细胞克隆形成率显著被抑制。放射线显著上调了A549细胞内Cbl-b蛋白表达。过表达Cbl-b显著降低A549细胞的增殖能力,并且放射敏感性显著升高,同时过表达Cbl-b通过下调c-Met增加A549细胞的放射敏感性。结论泛素连接酶Cbl-b通过下调c-Met提高肺癌细胞A549的放射敏感性。Objective To investigate whether ubiquitin ligase Cbl-b affects radiosensitivity of lung cancer by regulating c-Met.Methods The cells A549 were irradiated with radiation of 0 Gy,4 Gy,8 Gy,and 12 Gy.The absorbance value was measured by CCK-8 method,and the cell viability was calculated.Radioimmunoassay was detected by cell clone formation assay,and the expression level of ubiquitin ligase Cbl-b protein was detected by Western blot.A blank control plasmid(Control group)and a Cbl-b overexpression plasmid(Cbl-b group)were transfected into the A549 cells.Radiosensitivity was detected by CCK-8 and cell clone formation assays,and c-Met protein expression levels were detected by Western blot.Results with increasing dose.Radiation also inhibited the colony formation rate of A549 cells,and the cell clone formation rate was significantly inhibited as the radiation dose increased.Radioactivity significantly up-regulated the expression of Cbl-b protein in A549 cells.Overexpression of Cbl-b significantly reduced the proliferative capacity of A549 cells,and the radiosensitivity was significantly increased,while overexpression of Cbl-b increased the radiosensitivity of A549 cells by down-regulating c-Met.Conclusion The ubiquitin ligase Cbl-b increases the radiosensitivity of lung cancer cells A549 by down-regulating c-Met.
