摘 要:目的探讨富氢水改善感染性休克大鼠肠系膜血管反应性的作用机制。方法SD大鼠随机分为假手术组(sham)、感染性休克(Septic shock,SS)、富氢水(Hydrogen-rich water,HRS)。分别于休克后1h、2h、3h、4h、6h取肠系膜上动脉(superior mesenteric artery,SMA)制成血管环测定舒张反应性及钙敏感性变化;HE染色观察肠系膜上皮损伤;ELISA检测血浆中炎症因子(TNF-α、IL-1β、IL-6)水平;Western blot检测RhoA/Rho信号通路相关蛋白的表达。结果感染性休克发生2h后SMA血管反应性和钙敏感性逐渐降低,而HRS可以提升SS大鼠SMA血管反应性和钙敏感性;同时HRS可以改善SS大鼠肠粘膜上皮的病理损伤,抑制炎症反应,降低大鼠血浆中TNF-α、IL-1β及IL-6的水平;其调控机制与HRS激活RohA蛋白,促进MYPT-1磷酸化相关。结论HRS能调节SS大鼠SMA血管反应性,缓解肠系膜损伤,该作用可能与激活RhoA/Rho信号通路调节钙敏感性有关。Objective To investigate the mechanism of hydrogen-enriched water in improving mesenteric vascular reactivity in septic shock rats.Methods SD rats were randomly divided into sham group,septic shock(SS),and hydrogen-rich water(HRS).The superior mesenteric artery(SMA)was taken at 1 h,2 h,3 h,4 h,and 6 h after shock to measure diastolic reactivity and changes in calcium sensitivity;HE staining was used to observe mesenteric epithelial damage;ELISA was used to detect plasma inflammation Factor(TNF-α,IL-1β,IL-6)levels;Western blot was used to detect the expression of RhoA/Rho signaling pathway related proteins.Results Two hours after the occurrence of septic shock,SMA vascular reactivity and calcium sensitivity gradually decreased,and HRS could increase the SMA vascular reactivity and calcium sensitivity of SS rats,improve the pathological damage of intestinal mucosa of SS rats and inhibit inflammation response,reduce the levels of TNF-,IL-1βand IL-6 in plasma;its regulatory mechanism is related to HRS activation of RohA protein and promotion of MYPT-1 phosphorylation.Conclusion HRS can regulate SMA vascular reactivity and alleviate mesenteric damage in SS rats,which is related to activation of RhoA/Rho signaling pathway to regulate calcium sensitivity.
