摘  要:将源自鲨鱼软骨的高纯度硫酸软骨素和胶原蛋白以质量比2∶1配伍,得到鲨鱼硫酸软骨素胶原蛋白复方(SCC),并考察了该复方对骨关节炎(OA)的改善作用。在实验的第0、2、4、6天向小鼠膝关节腔定量注射Ⅱ型胶原蛋白酶,构建骨关节炎小鼠实验模型。同时每天分别以300、150 mg·kg^-1剂量灌胃小鼠给予SCC干预,持续4周。之后,取关节组织,测定肿胀程度,病理切片染色分析软骨损伤情况;通过酶联免疫吸附法(ELISA)检测血清IL-1β和IL-4表达水平。关节病理切片H&E染色结果显示,SCC 300 mg·kg^-1剂量对软骨细胞的修复优于150 mg·kg^-1剂量;Masson和天狼猩红染色结果显示,SCC 300 mg·kg^-1剂量补充胶原蛋白的能力高于150 mg·kg^-1剂量;番红-O染色结果显示,SCC 300 mg·kg^-1剂量对软骨的修复效果强于150 mg·kg^-1剂量;ELISA检测结果显示,SCC 300 mg·kg^-1剂量能显著降低血清中IL-1β水平,提升IL-4的水平。以上实验结果表明,SCC可恢复关节软骨中流失的胶原蛋白,降低IL-1β和上调IL-4的表达水平,从而显著改善Ⅱ型胶原蛋白酶所导致的小鼠膝关节炎症状,说明这种源自鲨鱼软骨的复方具有开发成膳食补充剂的良好前景。Chondroitin sulfate and collagen complex(SCC)was prepared by mixing high-purity chondroitin sulfate and collagen derived from shark cartilage in a ratio of 2∶1(w/w),and the effect of SCC on osteoarthritis(OA)was investigated.In the present study,we repeatedly performed quantitative injection of type II collagenase into the knee joint cavity of mice on day 0,2,4,6 of the experiment.During the construction of the OA model,mice were intragastrically administered with SCC at doses of 300,150 mg·kg^-1 per day for 4 weeks.After that,the cartilage damage was evaluated by histopathological section analysis and serum inflammatory cytokine level analysis.H&E staining of pathological sections showed that the effect of high-dose(300 mg·kg^-1)SCC was better than that of low-dose(150 mg·kg^-1)in repairing the chondrocytes.The results of Masson and Sirius red staining showed that the collagen supplementation ability of high-dose SCC was higher than that of low-dose.Safranine-O staining results showed that high-dose SCC had better cartilage repair effect than low-dose.ELISA analysis results showed that high-dose SCC could significantly reduce the IL-1βlevel and enhance the IL-4 level in serum.The above experimental results showed that SCC could significantly improve the symptoms of knee osteoarthritis in mice caused by type II collagen enzyme by reducing IL-1βlevel or promoting IL-4 level and recovering the lost collagen in articular cartilage,suggesting that this shark-derived complex had a good prospect of being developed into a dietary supplement.
