SIVA1基因在胃癌组织中的表达变化及与化疗药物敏感性的关系

摘　　要：目的观察SIVA1基因在胃癌组织中的表达变化,并分析其与化疗药物对胃癌组织抑制率的关系。方法采用免疫组化法检测60例份胃癌及60例份胃正常组织中SIVA1,采用体外组织三维培养法检测顺铂(CDDP)、5-氟尿嘧啶(5-FU)、多柔比星(ADM)对30例份胃癌组织(以SIVA1表达强度为中位值分为相对高表达组14例份和相对低表达组16例份)的抑制率。结果胃癌和胃正常组织中SIVA1表达阳性率分别为25.0%(15/60)、88.3%(53/60),两者比较,P<0.05。CDDP、5-FU、ADM对胃癌组织的抑制率分别为28.81%、28.20%、25.45%;以SIVA1表达强度为中位值将30例份胃癌组织分为相对高表达组(14例份)和相对低表达组(16例份),CDDP、5-FU、ADM对相对低表达组的抑制率分别为23.40%±1.91%、30.94%±3.94%、29.20%±2.23%,三种药物对相对高表达组的抑制率分别为34.59%±2.11%、26.71%±5.29%、25.44%±2.39%,两组CDDP抑制率比较,P均<0.05。结论SIVA1在胃癌组织中呈低表达,HDRA法能有效检测化疗药物在体外对胃癌组织的抑制效应,胃癌组织中SIVA1表达与CDDP对胃癌组织的抑制率呈正相关,SIVA1可作为评价CDDP对胃癌治疗效果的指标。Objective To detect the expression changes of SIVA1 in the gastric cancer tissues,and to analyze its relationship with the inhibitory rate of chemotherapy drugs on gastric cancer tissues.Methods The expression of SIVA1 in 60 cases of gastric cancer tissues and 60 cases of normal tissues was detected by immunohistochemical method.The histoculture drugs response assay was carried out to determine the inhibition rates of cisplatin(CDDP),fluorouracil(5-FU),and adriamycin(ADM)to 30 cases of gastric cancer tissues,and these 30 cases of gastric cancer tissues were divided into the relatively high expression group(14 cases)and the relatively low expression group(16 cases)according to the expression intensity of SIVA1.Results The positive rate of SIVA1 expression in the gastric cancer tissues and normal tissues was 25.0%(15/60)and 88.3%(53/60),respectively(P<0.05).The inhibition rates of CDDP,5-FU and ADM to gastric cancer tissues were 28.81%,28.20%,and 25.45%,respectively.The inhibition rates of CDDP,5-FU,ADM to the low expression group were 23.40%±1.91%,30.94%±3.94%,and 29.20%±2.23%,respectively,while the inhibition rates of these three drugs to the high expression group were 34.59%±2.11%,26.71%±5.29%,and 25.44%±2.39%,and significant difference was found in the inhibition rate of CDDP to gastric cancer between the two groups(P<0.05).Conclusions SIVA1 is low expressed in the gastric cancer tissues,and HDRA method can effectively detect the inhibitory effect of chemotherapy drugs on gastric cancer tissues in vitro.SIVA1 expression in the gastric cancer tissues is positively correlated with the inhibitory rate of CDDP on gastric cancer tissues,and SIVA1 may be served as an indicator to evaluate the therapeutic effect of CDDP on gastric cancer.
