摘　　要：目的探讨6-磷酸果糖激酶-2/果糖双磷酸酶-2同工酶3(PFKFB3)敲减对肺腺癌细胞A549增殖和转移的影响。方法将人肺腺癌细胞系A549分3组,si-ctrl转染阴性对照序列组、si-PFKFB3#1组和si-PFKFB3#2组,分别转染2种不同的敲减siRNA序列。WB检测PFKFB3蛋白表达,MTT检测细胞活力,流式细胞术检测细胞周期,transwell检测细胞转移能力。结果与si-ctrl组比较,si-PFKFB3#1组(0.103±0.024)和si-PFKFB3#2组(0.106±0.028)PFKFB3/GAPDH蛋白相对表达量显著降低(P<0.05);孵育48、72和96 h,si-PFKFB3#1组和si-PFKFB3#2组细胞活力降低,细胞增殖抑制率升高(P<0.05);si-PFKFB3#1组[(63.51±3.87)%]和si-PFKFB3#2组[(61.26±3.66)%]细胞G1期比例增加,si-PFKFB3#1组[(25.64±2.71)%]和si-PFKFB3#2组[(24.79±2.59)%]S期比例减少,差异均有统计学意义(P<0.05)。与si-ctrl组(305±27)比较,si-PFKFB3#1组(125±20)和si-PFKFB3#2组(136±18)转移细胞数显著降低(P<0.05)。结论PFKFB3敲减可以抑制肺腺癌细胞A549增殖和转移。Objective To investigate the effect of 6-phosphate fructose kinase-2/fructose bisphosphatase-2 isoenzyme 3(PFKFB3)knockdown on the proliferation and metastasis of lung adenocarcinoma A549 cells.Methods A549 cells were divided into three groups.The si-ctrl group was transfected with negative control sequence.The si-PFKFB3#1 group and si-PFKFB3#2 group were respectively transfected with two different siRNA sequences.PFKFB3 expression,cell viability,cell cycle and metastatic ability were detected by Western blot,MTT assay,flow cytometry and transwell assay,respectively.Results The PFKFB3/GAPDH ratio in si-ctrl group was higher than that in si-PFKFB3#1 group(0.103±0.024)and si-PFKFB3#2 group(0.106±0.028)(P<0.05).After treatment for 48,72 and 96 hours,compared with si-ctrl group,cell activity decreased and proliferative inhibition rate increased in si-PFKFB3#1 group and si-PFKFB3#2 group.Furthermore,compared with si-ctrl group,the proportion of cells in G1 phase increased and the proportion of cells in S phase decreased in si-PFKFB3#1 group[(63.51±3.87)%and(25.64±2.71)%,respectively]and si-PFKFB3#2 group[(61.26±3.66)%and(24.79±2.59)%,respectively](P<0.05).In addition,the number of metastatic cells in si-ctrl group(305±27)was higher than that in si-PFKFB3#1 group(125±20)and si-PFKFB3#2 group(136±18)(P<0.05).Conclusion PFKFB3 knockdown can inhibit the proliferation and metastasis of lung adenocarcinoma A549 cells.
