摘 要:将对单磷酸肽选择能力较强的二氧化钛(TiO2)和对多磷酸肽具有特异性作用的氢氧化镓氧化物(GaO(OH))掺杂在一起,结合到具有较大比表面积的还原石墨烯(rGR)表面,通过水热法制备出rGRTiO2-GaO(OH)(rGTG)复合材料。以该材料作为富集材料,建立了磷酸肽的MALDI-TOF检测方法。通过标准品和混合肽段样品评价了方法的检测性能,当牛血清白蛋白与β-酪蛋白(1000:1,V:V)的混合水解液通过rGTG富集后仍能清晰地检测到β-酪蛋白磷酸肽,表明该方法具有很高的灵敏度。使用牛奶样品对该方法进行验证,经过r GTG富集后能够检测到更多的磷酸肽,说明该方法对实际样品具有很好的检测效果。In this paper,titanium dioxide(TiO2),which has a strong selectivity for monophosphate peptides,and gallium hydroxide oxide(GaO(OH)),which has a specific affinity on polyphosphate peptides,were doped and combined with reduction graphene(rGR)with a large specific surface by hydrothermal method to prepare the rGR-TiO2-GaO(OH)(r GTG)composite material.A MALDI-TOF detection method for phosphopeptides was established by using r GTG as enrichment material.The detection performance of the method was evaluated by standard casein peptide and mixed peptide samples.The experimental results showed thatβ-casein phosphopeptides in mixed hydrolysate of bovine serum albumin andβ-casein(1000∶1,V∶V)could still be clearly detected after rGTG enrichment,indicating that the method had high sensitivity in phosphopeptides detection.Milk samples were employed to verify the method,and the results showed that more phosphopeptides were detected after rGTG enrichment,suggesting that the method had great effect on the detection of phosphopeptides in actual samples.
