摘　　要：目的探讨miRNA-486、miRNA-200、miRNA-10b在肺癌患者与非肺癌患者血液中表达的差异性,并分析其与肺癌遗传易感性的关联性。方法采用病例对照研究方法,选择海南医学院附属海南医院2012年1月至2016年7月期间呼吸内科肺癌患者93例作为病例组,非肺癌患者41例作为对照组,获得知情同意后,采集研究对象的血液样本用于提取总miRNA,采用real-time PCR法检测miRNA-486、miRNA-200、miRNA-10b的相对表达水平,并运用秩和检验方法对实验结果进行多组间分析。结果病例组与对照组患者的性别、年龄、吸烟史,饮酒史比较,差异均无统计学意义(P>0.05);病例组患者的血液miRNA-486相对表达水平明显低于对照组,而miRNA-10b相对表达水平明显高于对照组,差异均具有统计学意义(P<0.05),而两组间的miRNA-200相对表达水平比较差异无统计学意义(P>0.05);病例组中不同性别、年龄、吸烟史,饮酒史患者的miRNA-486、miRNA-200、miRNA-10b相对表达水平比较差异均无统计学意义(P>0.05)。结论miRNA-486和miRNA-10b与肺癌遗传易感性具有关联,有可能作为肺癌的遗传易感性生物标志物。Objective To investigate the expression differences of miRNA-486,miRNA-200,and miRNA-10b in the blood of lung cancer patients and non-lung cancer patients,and analyze their association with genetic susceptibility to lung cancer.Methods A total of 93 patients with lung cancer,who admitted to the Affiliated Hospital of Hainan Medical College from January 2012 to July 2016,were selected as the case group.And 41 patients with non-lung cancer were used as the control group.After obtaining informed consent,blood samples from the study subjects were collected for total miRNA extraction.Real-time PCR was used to detect the relative expression levels of miRNA-486,miRNA-200,and miRNA-10b.The results were analyzed by rank sum test.Results There was no significant difference in gender,age,smoking history and drinking history between the case group and the control group(all P>0.05);the relative expression level of miRNA-486 in the case group was significantly lower than that in the control group,while the relative expression level of miRNA-10b was significantly higher than that in the control group,with statistically significant differences(P<0.05).There was no significant difference in the relative expression level of miRNA-200(P>0.05);In the blood group,miRNA-486,miRNA-200,and miRNA-10b were also no significant difference in gender,age,smoking history and drinking history(P>0.05).Conclusion miRNA-486 and miRNA-10b are associated with genetic susceptibility to lung cancer,could be used as genetic susceptibility biomarkers for lung cancer.
