摘  要:目的:通过研究中医五行音乐对抑郁模型大鼠的行为学,以及大鼠海马组织中NLRP3和IL-1β蛋白表达的影响,对中医理论指导下五行音乐疗法治疗抑郁症的分子机制进行探讨。方法:50只SD大鼠,随机分为正常组、模型组、氟西汀组、音乐治疗组和音乐+氟西汀组,每组10只。除正常组外,其余各组的大鼠边造模边治疗,抑郁症大鼠模型采用随机慢性温和刺激建立,同时氟西汀组大鼠灌胃氟西汀30 mg/kg,灌胃量为15 mL/kg;音乐治疗组大鼠在治疗时置于一安静避光的房间,对其播放五行音乐中的宫调和羽调音乐;音乐+氟西汀组大鼠在灌胃氟西汀的同时进行宫调和羽调音乐治疗。大鼠的抑郁状态变化采用糖水偏好实验、旷场实验、强迫游泳实验等行为学实验进行评价,采用Western blotting法检测大鼠海马组织中NLRP3和IL-1β蛋白表达水平。结果:与正常组比较,模型组大鼠糖水偏好率、旷场实验中大鼠水平和垂直运动次数、强迫游泳时间等均明显减少(P<0.01);与模型组比较,各治疗组大鼠糖水偏好率、水平和垂直运动次数、强迫游泳时间均显著增加(P<0.05或P<0.01)。Western blotting结果显示,模型组大鼠海马组织中NLRP3和IL-1β蛋白表达明显高于正常组(P<0.01);各治疗组大鼠NLRP3和IL-1β蛋白表达均明显低于模型组,差异有统计学意义(P<0.05或P<0.01)。结论:五行音乐治疗对抑郁症模型大鼠有一定治疗效果,其治疗机制可能为影响海马NLRP3和IL-1β蛋白表达。Objective: To explore the molecular mechanism of five element music therapy in treating depression by studying the behavior of depression model rats and the expression of NLRP3 and IL-1β protein in hippocampus of rats. Methods: A total of 50 SD rats were randomly divided into normal group, model group,fluoxetine group, music therapy group and music +fluoxetine group, with 10 rats in each group. The rats were treated with modeling and treatment except for the normal group. The depression rat model was established with random chronic mild stimulation. At the same time, the positive drug group was given fluoxetine 30 mg/kg by gavage and the gavage was 15 mL/kg. The rats in the music therapy group were placed in a quiet and dark room during treatment, and played uterine and plume music of the five elements music were played. The music+fluoxetine group was given the uterine and feather music while gavage fluoxetine treatment. The changes of depression state in rats were evaluated by sugar water preference test, open field test, forced swimming test and other behavioral tests. The expression levels of NLRP3 and IL-1β protein in hippocampus tissue of rats were detected by Western blotting. Results: Compared with the normal group, the sugar preference rate, horizontal and vertical movement times and forced swimming time of rats in the model group were significantly reduced(P <0.01). Compared with the model group, the sugar preference rate, horizontal and vertical exercise times and forced swimming time of rats in each treatment group were significantly increased(P<0.05 or P<0.01). Western blotting results showed that the expression of NLRP3 and IL-1β protein in hippocampus of model group was significantly higher than that of normal group(P<0.01), and the expression of NLRP3 and IL-1β protein in each treatment group was significantly lower than that in model group(P<0.05 or P<0.01).
