摘 要:目的观察丙戊酸钠对氯化锂-匹罗卡品癫痫鼠模型海马区B1及B2激肽受体(KB2R)表达的影响并探讨其作用机制。方法健康Wistar大鼠35只,采用氯化锂-匹罗卡品腹腔注射法制作癫痫模型,癫痫模型干预组15只,分为急性期丙戊酸钠干预组(12 h,Ⅰ组)、静止期丙戊酸钠干预组(5 d,Ⅱ组)、慢性期丙戊酸钠干预组(60 d,Ⅲ组),每组5只;癫痫模型无干预组15只,分为急性期无干预组(12 h,Ⅰa组),静止期无干预组(5 d,Ⅱa组),慢性期无干预组(60 d,Ⅲa组),每组5只;设置空白对照组5只(Ⅳ组)。丙戊酸钠均经鼻饲管给药,在干预后的相应时间点处死动物取海马标本,应用逆转录聚合酶链反应(RT-PCR)及western blot方法分别检测各组动物不同时间点海马区的KB1R及KB2R的表达变化,并相互进行比较。结果给予丙戊酸钠干预后,Ⅰ组与Ⅰa组比较,Ⅰ组KB1R mRNA及蛋白质表达均显著下调,差异具有显著性(P<0.05),Ⅰ组KB2R mRNA及蛋白质表达均显著上调,差异具有显著性(P<0.05);Ⅱ组与Ⅱa组比较,Ⅱ组KB1R mRNA及蛋白质表达显著下调,差异具有显著性(P<0.05);Ⅲ组与Ⅲa组比较,Ⅲ组KB2R mRNA及蛋白质表达均显著上调,差异具有显著性(P<0.05);癫痫模型无干预组与空白对照组比较,Ⅰa组及Ⅱa组KB1R及KB2R及蛋白质表达均显著上调,差异具有显著性(P<0.05),Ⅲa组KB2R及蛋白质表达均显著上调,差异具有显著性(P<0.05)。结论KB1R在癫痫形成过程中对脑组织可能具有兴奋性细胞毒作用,KB2R可能对海马组织有保护作用,丙戊酸钠抗癫痫的干预实验也支持这个观点。Objective To investigate the changes and the possible mechanisms in the expressions of the kinin B1 and B2 receptors in the hippocampus of rats submitted to pilocarpine model of epilepsy.Methods Thirty-five healthy Wistar rats were randomly divided into six groups,that is the model groupsⅠgroup,Ⅱgroup,Ⅲgroup,and intrapulmonary injection of saline water control groupsⅠa group,Ⅱa group,Ⅲa group,after succession of 15 rats to kindle to establish the model of epilepsy by pilocarpine.To collect hippocampus tissues after the rats were to put to death,and to compared the expression levels of kinin B1 and B2 receptor mRNA by reverse transcriptase polymerase chain reaction(RT-PCR)in the hippocampus of rats.Results There were more significant upregulation of kinin B1 receptor mRNA expression(P<0.05)in theⅠand theⅡgroups and there was no alteration inⅢgroup.The expressional levels of B2 receptor mRNA were upregulated in theⅠ,ⅡandⅢgroups.Conclusion An altered balance between the kinin B1 and B2 receptor may play an important role in the onset and maintenance of epilepsy.
