摘 要:随着分子生物技术的迅速发展,物种分化的过程及原因成为进化生物学和生态学研究的热点之一。黄腰响蜜䴕(Indicator xanthonotus)是中国分布的唯一的响蜜䴕科鸟类,其系统发生关系和分化时间尚不清楚。本文运用细胞色素c氧化酶I亚基(COI)和细胞色素b(Cytb)基因作为分子标记,结合NCBI获取到的19个物种的COI和Cytb基因序列,采用最大似然法及贝叶斯法构建系统发育树,探讨了黄腰响蜜䴕的系统发育及起源的问题,通过分子定时的方法估算黄腰响蜜䴕的分化时间。结果表明:黄腰响蜜䴕与响蜜䴕科其余3个物种聚为一支,是此科中最先分化出来的物种,位于响蜜䴕科分支的基部;黄腰响蜜䴕与鳞喉响蜜䴕(I.variegatus)类群的遗传距离最小,与小响蜜䴕(I.exilis)类群的遗传距离最大;黄腰响蜜䴕的分化时间可以追溯到中新世阶段(时间节点为20.44 Ma)。Rapid development of molecular biotechnology has led to advances in evolutionary biology and ecology,including study of the process of species divergence.Yellow-rumped honeyguide(Indicator xanthonotus)is the only honeyguide species recorded in China,and its molecular phylogeny and divergence time are unclear.We selected cytochrome c oxidase subunit I(COI)and cytochrome b(Cytb)gene as molecular markers.We studied these markers in combination with the sequences of 19 species from NCBI,and used maximum likelihood methods and Bayesian Inference methods to construct a phylogenetic tree.We explored the phylogeny and origin of the yellow-rumped honeyguide,and evaluated the divergence time by molecular timing.The results showed that the yellow-rumped honeyguide and the remaining 3 species of Indicatoridae were clustered as one branch.Yellow-rumped honeyguide was the first species to diverge in this family,and is found at the basal position of this clade.Through the analysis of genetic distance,yellow-rumped honeyguide was considered to be the most closely related species to scaly-throated honeyguide(I.variegatus),and the farthest related species to least honeyguide(I.exilis).The divergence of the yellow-rumped honeyguide could be traced to the Miocene stage(time node was 20.44 Ma).
