摘 要:目的利用蛋白质组学的方法对唾液腺腺样囊性癌(SACC)患者癌组织标本进行特异性标记物鉴定,以寻找SACC诊断潜在的特异性标记物。方法通过双向荧光差异凝胶电泳技术检测4例SACC组织与癌旁正常腮腺组织差异表达蛋白点,应用质谱技术对重复性较好的蛋白进行鉴定。结果通过双向荧光差异凝胶电泳技术检测到SACC组织与癌旁正常腮腺组织差异表达的蛋白点39个,对这些差异表达蛋白点进行配对t检验(Average ratio≥1.5,P<0.05),得到表达上调的差异蛋白点24个,表达下调的差异蛋白点15个。对重复性较好的差异蛋白点进行质谱分析得到了5种蛋白质,分别为α-淀粉酶1、葡萄糖调节蛋白(GRP)、蛋白二硫键异构酶(PDI)、LASP1蛋白和PFN1蛋白,并且其表达量均升高。结论α-淀粉酶1、GRP、PDI、LASP1蛋白和PFN1蛋白可能为SACC早期诊断潜在的组织特异性标记物。Objective\To identify specific markers in cancer tissue samples from patients with salivary adenoid cystic carcinoma(SACC)using proteomics methods,and to find potential specific markers for the diagnosis of SACC.\Methods\Two-dimensional difference gel electrophoresis(2D DIGE)was used to detect differentially expressed protein spots between SACC tissue and adjacent normal parotid gland tissue in four cases of SACC,and mass spectrometry was used to identify the proteins with good reproducibility.\Results\A total of 39 differentially expressed protein spots between SACC tissue and normal parotid gland tissue were detected by 2D DIGE.The paired t-test(Average ratio≥1.5,P<0.05)was performed on these differentially expressed protein spots to find 24 spots with up-regulated expression and 15 spots with down-regulated expression.Five proteins,namelyα-amylase 1,glucose regulated protein(GRP),protein disulfide isomerase(PDI),LASP1 protein,and PFN1 protein,were identified by mass spectrometry analysis of the differentially expressed protein spots with good reproducibility.\Conclusion\α-Amylase 1,GRP,PDI,LASP1 protein,and PFN1 protein may be potential tissue-specific biomarkers for early diagnosis of SACC.
