摘 要:真菌毒素是真菌产生的一类具有生物体内蓄积效应的有害次生代谢物,其广泛存在于食品、饲料及中药材中,对人体和动物具有潜在危害。因此,掌握体内真菌毒素准确、快速、高效的检测前沿技术,对真菌毒素致病机制探索及人体暴露评估具有重要意义。本文归类分析了常见单一真菌毒素如曲霉属真菌毒素、镰刀菌属真菌毒素的毒性效应及混合真菌毒素的联合毒性;剖析了尿液、血液、组织中真菌毒素及其标志物的样品制备和检测方法,如蛋白质沉淀、液液萃取、固相萃取、免疫亲和净化、直接稀释等生物样本的前处理;系统性介绍了真菌毒素毒性及其在生物样本的检测技术,如薄层色谱法、高效液相色谱法、色谱-质谱联用法、免疫分析法及其他如适配体传感器技术、噬菌体展示技术和胶体金技术等。本综述可为体内真菌毒素及其标志物检测研究和人体真菌毒素暴露风险评估提供参考。Mycotoxins are a class of harmful secondary metabolites produced by fungi that have a bioaccumulative effect.They are widely found in foods,feeds,and Chinese medicinal materials.They can enter the human food chain from daily diet and cause great harm to humans and animals.Therefore,accurate,rapid and effective detection of mycotoxin in vivo is of great significance for the detection of pathogenic mechanisms and the assessment of human exposure.This paper classifies and analyzes the toxic effects of single mycotoxins such as Aspergillus mycotoxin and Fusarium mycotoxin and the combined toxicity of mixed mycotoxins.Pre-treatment methods and detection techniques for mycotoxins and their markers in urine,blood samples and tissues are described,such as the purification methods of biological sample pretreatment include protein precipitation,liquid-liquid extraction,solid phase extraction,immunoaffinity purification,and direct dilution.The toxicity of mycotoxins and their detection techniques in biological samples,such as TLC,HPLC,GC-MS,immunoassay and other methods,such as aptamer sensor technology,phage display technology and colloidal gold technology,were introduced.This review aims to provide reference for the detection of mycotoxins and their biomarkers in vivo and risk assessment of human mycotoxins exposure.
