摘  要:目的通过行为学测试及病理学观察帕金森病(PD)大鼠黑质区形态学变化,对帕金森病大鼠模型进行评价,探讨PD大鼠行为学变化的病理学基础并为后期运用功能核磁共振观察PD老鼠脑区功能连接的改变情况提供实验基础。方法将60只健康雄性SD大鼠随机分为正常组和模型组各30只,采用6-羟基多巴胺(6-OHDA)制作大鼠模型,分别于术后7d、14d、21d腹腔注射阿朴吗啡(APO)检测其旋转行为,转圈数≥210r/30min的大鼠为成功的帕金森病实验模型。通过旷场实验和露台水迷宫实验评价模型组及正常组大鼠的行为学差异;光镜下观察模型组及正常组大鼠脑黑质神经元数量的变化情况。结果采用内侧前脑束单侧两点注射6-羟基多巴胺(6-OHDA)的方法成功建立PD大鼠模型。旷场实验结果显示模型组大鼠直立次数和运动总距离低于正常组;露台水迷宫实验中模型组大鼠寻台时间延长;HE染色下,模型组大鼠单侧黑质区神经元数量减少。结论采用内侧前脑束单侧两点注射6-OHDA建立PD大鼠模型的方法高效、稳定,PD大鼠的行为学变化有其病理学基础。Objective To observe the morphological changes of the substantia nigra in the rats with Parkinson’s disease(PD) by behavioral test and pathology, and the model of PD was evaluated to explore the pathological basisof the behavioral changes of PD rats and provide experimental basis for the use of fMRI to observe the changes of functional connections in the brain of PD rats.Method Sixty SD rats were randomly divided into two groups:normal group(n=30),and experimental group(n=30).The rat model was made with 6-OHDA. Apomorphine(APO) was injected intraperitoneally on 7 d, 14 d and 21 d after operation to detect the rotation behavior.The successful experimental model of Parkinson’s disease was established in rats with rotation ≥ 210 r/30 min.Evaluation of behavioral differences was made between model group and normal group by open field experiment and terrace water maze experiment. The number of neurons in the substantia nigra of the model group and the normal group was observed under the light microscope.Result Twenty successful models were chosen as the model group.The results of open field experiment showed that the number of standing up and the total distance of movement in the model group were lower than those in the normal group and the model group rats needed more time to reach the terrace morris water maze experiment. HE staining result revealed that the number of neurons in the unilateral substantia nigra decreased in the model group.Conclusion The experimental resuts showed that the unilateral injecting of 6-OHDA into the medial forebrain bundle was an effective and stable method. The behavioral changes of PD rats had its pathological basis.
