摘 要:目的揭示肺腺癌中磷酸核糖甘氨酰胺转甲酰酶(GART)基因变化及其与患者预后的关系。方法2020年1月使用癌症芯片数据库Oncomine、癌症基因组学分析门户网站cBioPortal、癌症基因组学门户网站UALCAN及生物信息学数据库DAVID 6.8分析肺腺癌组织GART基因变化及其与患者预后的关系。结果Oncomine数据库结果显示,肺腺癌组织GART基因表达明显升高。cBioPortal数据库结果显示,501例肺腺癌患者中发生GART基因改变38例,包括错义突变、截短突变、基因融合、拷贝数畸变和mRNA水平上调。GART基因高表达不利于患者预后,差异有统计学意义(P<0.05)。UALCAN数据库结果显示,肺腺癌组织GART基因表达明显提高,且在40岁以上、吸烟人群中GART的表达更为显著。DAVID富集分析结果显示,GART主要与细胞周期、RNA转运、DNA复制途径相关。结论肺腺癌组织GART高表达不利于患者预后,该基因可作为判断肺腺癌患者预后的候选标志物。Objective To reveal the relationship between the changes of glycinamide ribonucleotide transformylase(GART)gene and prognosis in the patients with lung adenocarcinoma(LUAD).Methods The cancer chip database Oncomine,cancer genomics analysis web portal cBioPortal,cancer genomics web portal UALCAN and the bioinformatics database DAVID 6.8 were used to analyze the changes of GART gene in LUAD tissues and its relationship with clinical prognosis of the patients in January 2020.Results The Oncomine database results showed that GART gene expression was significantly elevated in LUAD tissue.The cBioPortal database results showed that GART gene changes occurred in 38 cases among 501 LUAD patients,including missense mutations,truncation mutations,gene fusion,copy number aberrations and mRNA level upregulation.GART gene high expression of GART was not conducive to the patient′s prognosis,and the difference was statistically significant(P<0.05).The UALCAN database results showed that the expression of GART in LUAD tissues was significantly increased,moreover the expression of GART was more significant in the individuals aged over 40-year-old and smokers.The enrichment analysis of DSVID showed that GART was mainly related to the cell cycle,RNA transport and DNA replication pathway.Conclusion High expression of GART in LUAD tissue is unfavorable to the prognosis and can serve as a candidate marker for judging the prognosis of LUAD patients.
