摘 要:本试验以中国荷斯坦奶牛为研究对象,旨在通过筛选血管生成素样蛋白5(angptl5)基因的突变位点,获得影响中国荷斯坦奶牛乳品质性状的潜在遗传标记。PCR扩增angptl5基因的目的片段,通过直接测序的方法检测目的基因片段的单核苷酸多态性,使用SPSS 23.0软件分析其与中国荷斯坦奶牛乳品质性状的相关关系。结果发现,angptl5基因内含子中存在1个突变位点,在检测群体中该I1-1776G>A突变位点存在AA、AG、GG3种基因型,通过基因多态性分析,发现I1-1776G>A位点基因杂合度(H)和多态信息含量(PIC)均处于0.25~0.50之间,χ^2=1.85,P>0.05,说明I1-1776 G>A位点在中国荷斯坦奶牛群体中处于哈代温伯格平衡状态(P>0.05)。其中AA基因型个体的尿素氮显著高于AG、GG型(P<0.05),而与产奶量、乳蛋白、乳糖、脂肪以及干物质含量均无显著相关性(P>0.05)。研究结果可为未来研究奶牛angptl5基因生物学功能和优质奶牛选育提供分子理论依据。The purpose of this experiment is to obtain potential genetic markers that affect milk quality traits of Chinese Holstein dairy cows by screening for mutations in the angiogenin-like protein 5(ANGPTL5).The fragment of ANGPTL5 gene was amplified by PCR,and the single nucleotide polymorphism of the target gene fragment was detected by direct sequencing.The correlation between this single nucleotide polymorphism and milk quality traits of Chinese Holstein dairy cows was analyzed by SPSS 23.0 software.The results showed that there was one mutation site in the intron of ANGPTL5 gene,and there were three genotypes of AA,AG,and GG in the I1-1776 G>A mutation site in the test population.Through the analysis of gene polymorphism,it was shown that the heterozygosity(H)and(PIC)of I1-1776 G>A locus were in the range of 0.25-0.50,χ^2=1.85,P>0.05,indicating that I1-1776 G>A locus was in Hardy Weinberg equilibrium in Chinese Holstein dairy population(P>0.05),the content of urea nitrogen of AA genotype individuals is significantly higher than that of AG and GG types(P<0.05),but it is no significant correlation with milk production,milk protein,lactose,fat and dry matter content(P>0.05).The results of this study can provide molecular theoretical basis for future studies on the biological function of ANGPTL5 gene in dairy cows and the selection of high quality dairy cows.
