摘 要:目的建立小鼠抑郁症模型,通过激光散斑成像技术在体方法观察抑郁症小鼠皮层扩散性抑制(CSD)的脑血流动力学改变。方法采用社会隔离(SI)诱导慢性小鼠抑郁症模型,通过糖水偏好实验、旷场实验和悬尾实验检测造模后小鼠抑郁样行为学改变;利用激光散斑成像技术监测小鼠皮层扩散性抑制的血流动力学改变。应用免疫荧光方法对星形胶质细胞和小胶质细胞进行染色,计数并统计分析。结果社会隔离慢性抑郁症造模方法可引起小鼠抑郁样的行为改变。激光散斑成像技术监测抑郁症小鼠CSD过程中脑血流变化率明显小于正常小鼠(P<0.001),脑血流达峰时间延迟,充血持续时间延长(P<0.01)。星形胶质细胞在抑郁症小鼠中明显减少(P<0.05)。结论本实验方法成功构建小鼠慢性抑郁症模型;在体条件下激光散斑成像技术监测发现抑郁症小鼠脑血流的调节能力受损。Objectives Establish a mouse model for chronic depression disease and observe the hemodynamic changes in depression mice.Methods Social isolation(SI)was applied to induce chronic depressive.Tail suspension test,open field test and sucrose preference test were used to detect depression-like behaviors of mice.Laser speckle contrast imaging was applied to monitor the changes of cerebral blood flow dynamics after cortical spreading depression in mice.The astrocytes and microglia were visualized with immunofluorescence method,the number of the cells were counted and analyzed.Results SI could induce depression-like behaviors in mice.Laser speckle contrast imaging results showed that the change rate of CSD-induced cerebral blood flow of depression mice was notable lower than that of normal mice(P<0.001).Cerebral blood flow peak time was delayed,and hyperemia duration was prolonged in depression mice(P<0.01).The number of astrocytes were decreased significantly in chronic depressive mouse model(P<0.05).Conclusion SI could induce chronic depression and impair regulation of cerebral blood flow.
