高、低肥肝性能骡鸭肠道形态、菌群及其代谢物比较

摘  要:为了比较高、低肥肝性能的骡鸭肠道形态、菌群及菌群代谢产物的差异性,试验将同批次、相同饲养条件下、体重相近的70日龄公骡鸭300只进行填饲肥肝19 d后,屠宰骡鸭,根据肝重和肝体比(肝重与体重的比值)将试验鸭只分为高肥肝组(肝重大于850 g且肝体比大于13.9%)和低肥肝组(肝重小于380 g且肝体比小于6.9%),采集两组鸭只的盲肠和空肠制成组织切片;采用16S rDNA测序技术测定两组鸭只盲肠内容物菌群差异情况;采用GC-MS短链脂肪酸平台对两组骡鸭盲肠内容物短链脂肪酸进行定量检测分析。结果表明:高肥肝组骡鸭盲肠和空肠肠道形态结构比低肥肝组更完整;高、低肥肝组骡鸭盲肠菌群在门和属水平上无显著差异(P>0.05);高肥肝组骡鸭盲肠内容物中异丁酸、2-甲基丁酸、丁酸、异戊酸极显著高于低肥肝组(P<0.01),4-甲基戊酸极显著低于低肥肝组(P<0.01)。说明肠道内异丁酸、2-甲基丁酸、丁酸、异戊酸对骡鸭肥肝的形成具有促进作用,4-甲基戊酸对骡鸭肥肝的形成可能具有抑制作用。In order to compare the differences in intestinal morphology,flora and metabolites of mule duck with high and low liver production performance,300 mule ducks of similar weight in the same batch were fed fatty liver under the condition of the same breeding.After feeding for 19 days,the mule ducks were slaughtered and divided into a high-yielding group(the liver weight was more than 850 g and the liver-body ratio was more than 13.9%)and a low-yielding group(the liver weight was less than 380 g and the liver-body ratio was less than 6.9%)based on liver weight and liver-body ratio(liver-body weight ratio).The cecum and jejunum of two groups of ducks were collected for preparation of tissue sections.16 S rDNA sequencing technique was used to determine the differences of cecum contents flora between the high-yielding group and the low-yielding group.At the same time,GC-MS short chain fatty acid platform was used to quantitatively detect the short chain fatty acids in cecum contents.The results showed that the intestinal morphological structure of the high-yielding group was more complete than that of the low-yielding group.The detection of intestinal flora showed that there was no significant difference of intestinal flora in cecum at the level of phylum and genus(P>0.05).Isobutyric acid,2-methyl-butyric acid,butyric acid,and iso-valerate acid in the cecal contents of the high-yielding group were significantly higher than that of the low-yielding group(P<0.01),and 4-methyl-valerate acid were significantly lower than that of the low-yielding group(P<0.01).The results showed that intestinal iso-butyric acid,2-methyl-butyric acid,butyric acid,and iso-valeric acid could promote the formation of mule duck fatty liver,and 4-methyl-valeric acid may inhibit the formation of fatty liver of mule duck.
