摘 要:目的探讨DJ-1基因在非急性早幼粒细胞白血病(APL)的急性髓系白血病(AML)患者骨髓单个核细胞中的表达、临床特征和其对预后的影响。方法收集135例患者骨髓标本,采用实时荧光定量PCR检测DJ-1表达水平,分析其与临床特征及预后的相关性。结果在mRNA水平上,初治组与复发难治组的DJ-1表达水平明显高于正常对照组及缓解组(P<0.01);初治组与复发难治组的DJ-1表达水平无明显差异(P>0.05);缓解组DJ-1表达水平与对照组差异不明显(P>0.05);初治组中DJ-1的水平与白细胞计数高低有关(P<0.01),而与患者的年龄、性别、贫血程度、血小板计数高低、幼稚细胞高低、FAB分型、WHO分型无统计学差异(P>0.05)。结论DJ-1基因在初治及复发难治非APL的AML患者中存在高表达,缓解组表达下降,且初诊患者中DJ-1的表达与患者白细胞高低有相关性。Objective To explore the expression gene of DJ-1、clinical features in bone marrow mononuclear cells of Acute myeloid leukemia(AML)patients with non-acute promyelocytic leukemia(APL)and its effect on prognosis.Methods Bone marrow specimens were collected from 135 patients,and DJ-1 expression levels were detected by real-time quantitative PCR to analyze their correlation with clinical characteristics and prognosis.Results At the mRNA level,the expression of DJ-1 in the Initial treatment group and the relapsed and refractory group was significantly higher than that in the normal control group and the remission group(P<0.01);There was no significant difference in the expression of DJ-1 between the newly treated group and the relapsed and refractory group(P>0.05);the DJ-1 expression level of the patients in the remission group was not significantly different from that in the control group(P>0.05);The level of DJ-1 in the newly treated patient group was related to the level of white blood cell count(r=0.5281,P<0.0001),There was no significant difference with patients'age,gender,degree of anemia,platelet count,immature cell level,FAB classification,and WHO classification(P>0.05).Conclusion The DJ-1 gene is highly expressed in patients with newly diagnosed and relapsed and refractory AML with non-acute promyelocytic leukemia,The expression of patients in remission phase decreased,and the expression of DJ-1 in newly diagnosed patients was related to the level of white blood cells.
