摘 要:目的研究ATP柠檬酸裂解酶(ACLY)基因对结肠癌细胞脂质代谢、增殖、凋亡和侵袭的影响。方法体外培养HCT116结肠癌细胞,根据细胞处理方式分为3组:以敲减ACLY基因的HCT116细胞作为ACLY敲减组,以空载体转染的HCT116细胞作为阴性对照组,以未经处理的HCT116细胞作为空白对照组。以嘌呤霉素筛选出稳定的新细胞系后,采用蛋白质印迹法检测ACLY蛋白的表达情况;采用甘油三酯检测试盒检测细胞甘油三酯水平,采用CCK-8法检测细胞的增殖能力,采用流式细胞仪检测细胞凋亡情况,采用细胞划痕实验检测细胞的迁移能力。结果ACLY敲减组的细胞存活率在120 h时相较于空白对照组和阴性对照组降低,但在24 h和48 h时3组的细胞存活率比较差异不大;与阴性对照组和空白对照组比较,ACLY敲减组的凋亡率升高,24 h的迁移率以及胞内甘油三酯水平降低。结论ACLY基因敲减能够抑制结肠癌细胞的增殖、凋亡和迁移,其机制可能与细胞的脂质合成能力降低有关。Objective To study the effects of ATP citrate lyase(ACLY)gene on proliferation,apoptosis,invasion,and lipid metabolism of colon cancer cells.Methods Colon cancer cells HCT116 were transfected with lentiviral knockdown ACLY gene in vitro and divided into three groups according to cell treatment:HCT116 cells with ACLY gene knockdown as knockdown group,empty vector transfected cells as negative control group,and untreated colon cancer HCT116 cells as blank control group.After the stable new cell line was screened with puromycin,the expression of ACLY protein was detected by Western blot method,the lipid production of cells was detected by triglyceride test kit,the proliferation ability of cells was detected by CCK-8 method,the apoptosis rate was detected by flow cytometry,and the migration ability of cells was detected by cell scratch test.Results The cell survival rate of the knockdown group was lower than those of the blank control group and the negative control group at 120 h,but there was no significant difference among the three groups at 24 h and 48 h.Compared with the negative control group and the blank control group,the apoptosis rate in the knockdown group increased,the 24 h migration ability and the level of intracellular triglyceride decreased.Conclusion ACLY gene knockdown can inhibit the proliferation,apoptosis,and migration of colon cancer cells,and its mechanism may be related to the decrease of lipid synthesis ability of colon cancer cells.
