摘  要:目的探讨人类羟基类固醇脱氢酶样蛋白2(hydroxysteroid dehydrogenase like 2,HSDL2)在人头颈鳞状细胞癌(head and neck squamous cell carcinoma,HNSCC)中的表达及临床意义。方法通过UALCAN数据库分析HNSCC中HSDL2 mRNA的表达及其与临床病理学特征的关系;应用The Human Protein Atlas(HPA)数据库和免疫组化SP两步法染色检测HNSCC中HSDL2蛋白的表达,并分析其与临床病理学特征的关系;应用HPA数据库分析HSDL2 mRNA表达与HNSCC患者生存期的关系;应用STRING数据库分析HSDL2可能参与的生物学进程及蛋白互作网络。结果与癌旁正常组织相比,HNSCC组织中HSDL2 mRNA的表达水平明显降低(P<0.01)。免疫组化染色显示HSDL2蛋白主要定位于细胞质中,且在HNSCC组织中的表达明显低于癌旁正常组织(P<0.05);HSDL2低表达与临床分期(P<0.01)、淋巴结转移(P<0.05)和患者预后密切相关(P<0.05)。GO富集分析显示,HSDL2主要参与细胞的脂肪酸代谢过程。结论HSDL2低表达与HNSCC的恶性演进密切相关,提示HSDL2在HNSCC的发生、发展中发挥抑癌基因的作用,同时其表达水平对于HNSCC的早期诊断和预后评估具有一定的指导意义。Purpose To exlpore the expression and clinicopathological significance of hydroxysteroid dehydrogenase like 2(HSDL2)in head and neck squamous cell carcinoma(HNSCC).Methods UALCAN database was used to analyze the expression of HSDL2 mRNA and its relationship with clinicopathological parameters in HNSCC.The Human Protein Atlas(HPA)database and immunohistochemistry were performed to detect the expression of HSDL2 protein in HNSCC.The correlation between HSDL2 expression and clinicopathological features of HNSCC was analyzed.HPA database was applied to analyze the relationship between the expression of HSDL2 mRNA and survival time of HNSCC patients.The STRING database was utilized to analyze the proteins interacting with HSDL2 and the HSDL2-involved biological process.Results The expression of HSDL2 mRNA in HNSCC was significantly lower than that in adjacent normal tissues(P<0.01).Immunohistochemistry showed that HSDL2 was mainly located in the cytoplasm,and its expression in HNSCC was significantly lower than that in adjacent normal tissues(P<0.05).The low expression of HSDL2 protein was correlated with clinical stage(P<0.01),lymph node metastasis(P<0.05)and prognosis in HNSCC(P<0.05).The GO enrichment analysis indicated that HSDL2 was mainly involved in the process of fatty acid metabolism.Conclusion The low expression of HSDL2 is associated with the malignant progression of HNSCC,suggesting that HSDL2 may play a function of tumor suppressor gene in the initiation and progression of HNSCC,and its expression has a certain clinical significance for the early diagnosis and prognosis evaluation of HNSCC.
