摘 要:目的探讨咖啡因对小鼠皮肤伤口愈合和愈后瘢痕的影响。方法采用以往建立的通过饮用的方式摄取咖啡因慢性干预小鼠模型和小鼠背部全层皮肤切割伤模型,伤后每隔2 d采用透明膜描绘法记录伤口大小,愈后30 d记录瘢痕大小;HE染色观察伤后3、6、9 d和愈合后30 d的伤口和瘢痕组织学变化,并结合Labwork 4.0软件分析伤口组织上皮爬行、肉芽组织厚度、瘢痕组织上皮薄厚比及瘢痕宽度变化;Masson染色分析伤后6 d和愈合后30 d的伤口和瘢痕组织的胶原变化;采用免疫组化观察伤后6 d伤口组织CD31和CD11b蛋白水平变化。结果咖啡因组小鼠伤口愈合时间比对照组延长了28.3%,但是创伤后瘢痕面积比对照组减低了56.8%;伤口愈合过程中,咖啡因组不仅在上皮爬行长度、肉芽组织厚度都显著低于对照组(P<0.01),而且伤口中胶原含量、CD31表达强度降低(P<0.01),CD11b表达强度提高(P<0.01);愈后瘢痕中,咖啡因组小鼠瘢痕宽度要小于对照组(P<0.01),瘢痕组织上皮薄厚比大于对照组(P<0.01),而瘢痕中胶原染色较与周边未损伤皮下胶原染色相近。结论咖啡因可延缓小鼠皮肤伤口愈合,但可减小瘢痕形成。Objective To investigate the effect of caffeine consumption on skin wound healing and scar formation in mice. Methods The mouse model was established according to our previous method of chronic intervention with caffeine combined with the model of full-thickness skin wound on the back. Wound closure was traced every 2 days in each group using transparent film until the wound closed, and the scar areas were calculated at 30 d after healing. The sections were stained with hematoxylin-eosin(HE) to observe the changes in epithelial cells crawling and thickness of granulation tissue at 3, 6 and 9 d after wounding, and scar width and the thinness-to-thickness ratio of the epithelium in the scar tissue at 30 d after healing with the aid of Labwork 4.0 software. Masson staining was used to analyze the collagen changes of the wound at 6 d after wounding and of the scar tissue at 30 d after healing. Immunohistochemical assay was used to observe the changes of CD31 and CD11 b protein levels in wound tissue at 6 d after wounding. Results The healing time of caffeine group was delayed by 28.3%, and the scar area was reduced by 56.8% when compared with that of the control group. In the process of wound healing, the crawling length of epithelium and the thickness of granulation tissue were significantly decreased in the caffeine group than the control group(P<0.01), the collagen content and the expression level of CD31 in the wound were also decreased(P<0.01), and the expression level of CD11 b was obviously enhanced(P<0.01). Meanwhile, compared with the control group, the caffeine group had significantly decreased scar width(P<0.01), higher thickness of scar tissue epithelium(P<0.01), but similar collagen content in the scar and the surrounding undamaged subcutaneous collagen. Conclusion Caffeine consumption can delay the wound healing in mice, but attenuate scar formation.
