摘  要:目的:利用数据库和生物信息学的方法分析己糖激酶2(HK2)在泌尿系肿瘤中表达水平、预后及与肿瘤免疫细胞浸润的相关性.方法:研究HK2在癌组织和癌旁组织mRNA表达水平,其表达水平与肿瘤预后的相关性,分析HK2表达水平与肿瘤免疫细胞浸润、免疫细胞标志物表达水平、肿瘤微环境免疫细胞和基质细胞浸润的相关性.结果:HK2表达在泌尿系肿瘤组织中均显著性升高,在膀胱癌(BLCA)中随肿瘤分级增加而降低,在高级别肾乳头状细胞癌(KIRP)和前列腺癌(PRAD)中HK2表达水平显著高于低级别表达水平;BLCA和肾透明细胞癌(KIRC)中HK2低表达的患者总体生存率显著低于高表达患者,而KIRP中则相反.HK2表达水平与BLCA和KIRP中多数免疫细胞浸润水平无明显相关性,与KIRC和PRAD中多数免疫细胞浸润水平呈显著正相关.与免疫细胞标志物表达相关性分析发现,HK2在BLCA中表达与免疫细胞标志物表达呈显著负相关,而在KIRC、KIRP和PRAD中大多呈显著正相关.HK2在BLCA和KIRC中表达水平与肿瘤微环境呈显著正相关,在PRAD和KIRP中则无显著相关性.结论:HK2表达水平与BLCA、KIRC和KIRP患者的预后相关;HK2表达水平与免疫细胞浸润性、免疫细胞标志物表达、肿瘤微环境免疫细胞和基质细胞浸润等相关.Objective:Analysis of the correlation between expression level of hexokinase 2(HK2),prognosis and tumor immune cell infiltration in urinary tract tumors using database and bioinformatics methods.Methods:Analyzing the mRNA expression level of HK2 in cancer tissues and adjacent tissues,its correlation with tumor prognosis and tumor immune cell infiltration,the correlation between HK2 expression level and immune cell marker expression level,and correlation of tumor microenvironment immune cell and stromal cell infiltration.Results:HK2 expression was significantly increased in urinary tract tumor tissues.In BLCA,HK2 expression decreased with the increase of tumor grade.In high grade KIRP and PRAD,expression level of HK2 was significantly higher than that in low grade.The OS of patients with low-profile is significantly shorter than high-expression patients,while that is opposite for patients with KIRP.Expression level of HK2 had no significant correlation with the infiltration level of most immune cells in BLCA and KIRP.However,it positively correlated with the infiltration level of most immune cells in KIRC and PRAD.Analysis with the expression of immune cell markers found that the expression of HK2 in BLCA was significantly negatively correlated with the expression of immune cell markers,while most of them were significantly positively correlated in KIRC,KIRP,and PRAD.The expression level of HK2 in BLCA and KIRC was significantly positively correlated with tumor microenvironment yet there was no significant correlation in PRAD and KIRP.Conclusion:Expression of HK2 is associated with the prognosis of patients with BLCA,KIRC and KIRP.The expression level of HK2 is correlated to the infiltration of immune cells,the expression of immune cell markers,and the tumor microenvironment.
