摘　　要：目的:检测多聚嘧啶结合蛋白3(polypyrimidine tract binding protein 3,PTBP3)在胃癌组织中的表达及定位,探讨其表达水平与根治术后胃癌患者临床病理特征及生存预后的关系。方法:收集90例根治术后胃癌患者组织及对应的癌旁组织标本,制成组织芯片后,采用免疫组织化学(SP染色系统)法检测PTBP3蛋白表达及定位。收集30例配对胃癌及癌旁组织标本,制成cDNA芯片后,采用实时荧光定量PCR法检测PTBP3 mRNA的表达水平。应用SPSS 19.0统计学软件分析PTBP3表达与88例胃癌根治术后患者临床病理特征及生存预后的关系。结果:PTBP3主要定位在细胞核上,胃癌组织中PTBP3蛋白(细胞质P<0.001,细胞核P=0.011)和mRNA(P=0.036)的表达水平均明显高于癌旁组织。多因素分析显示,TNM分期(P=0.031)和癌旁组织中PTBP3表达(P=0.014)为影响根治术后胃癌患者总生存期(overall survival,OS)的独立预后因素;癌旁组织PTBP3蛋白低表达组的中位OS期较高表达组明显延长(68.58 vs 27.27个月,P=0.016)。结论:PTBP3在胃癌中可能具有癌基因的功能,其可能成为胃癌靶向治疗的潜在候选基因。Objective:To detect the expression and localization of polypyrimidine tract binding protein 3(PTBP3)in gastric cancer tissues,and to explore the relationships of PTBP3 expression level with the clinicopathological characteristics and survival prognosis of patients with radical gastric cancer.Methods:A total of 90 cases of gastric cancer tissues and the corresponding adjacent tissue samples from the patients after radical resection were collected to make tissue chips,then the expression and location of PTBP3 protein were detected by immunohistochemistry(SP staining system).Thirty matched samples of gastric cancer and the adjacent tissues were collected to make cDNA microarray,then the expression level of PTBP3 mRNA was detected by real-time fluorescent quantitative PCR.SPSS 19.0 statistical software was used to analyze the relationships of PTBP3 expression with the clinicopathological characteristics and survival prognosis of 88 patients after radical gastric cancer surgery.Results:PTBP3 was mainly located in the nucleus,and its protein(cytoplasm P<0.001,nuclear P=0.011)and mRNA(P=0.036)levels in gastric cancer tissues were significantly higher than those in the adjacent tissues.Multivariate analysis showed that TNM staging(P=0.031)and the expression level of PTBP3 protein in the adjacent tissues(P=0.014)were independent prognostic factors that affected the overall survival(OS)of gastric cancer patients after radical resection.The median OS in the low expression group of PTBP3 protein in the adjacent tissues was significantly prolonged as compared to that in the high expression(68.58 months vs 27.27 months,P=0.016).Conclusion:PTBP3 may have function as an oncogene in gastric cancer,and it may become a potential candidate gene for targeted therapy of gastric cancer.
