MRP1、GLUT1在骨肉瘤中的表达水平及其与临床病理特征的关系

摘　　要：目的:观察骨肉瘤多药耐药相关蛋白1(MRP1)、葡萄糖转运蛋白-1(GLUT1)的表达水平及其与临床病理特征的关系。方法:回顾性分析2015年10月至2016年10月60例骨肉瘤患者的临床资料,设研究组(60份骨肉瘤组织标本)及对照组(60份瘤旁正常骨组织标本,距肿瘤边缘>5 cm处)。两组标本均采用免疫组化法检测MRP1、GLUT1的表达水平,比较两组MRP1、GLUT1的表达水平并分析其与临床病理特征的关系。结果:研究组MRP1、GLUT1阳性表达率明显高于对照组,差异有统计学意义(P<0.05);临床分期为Ⅱb~Ⅲ期、肿瘤直径小、分化程度中低以及生存期<1年的骨肉瘤患者,MRP1、GLUT1蛋白阳性表达率分别高于临床分期为Ⅰ~Ⅱa期、肿瘤长径大于所在骨直径5倍以上、分化程度高以及生存期≥1年者,差异有统计学意义(P<0.05);MRP1、GLUT1阳性表达患者中位生存期明显短于阴性表达患者,差异有统计学意义(P<0.05)。结论:MRP1、GLUT1在骨肉瘤组织中呈高表达,且其水平与肿瘤的临床分期、直径大小、分化程度及生存期相关。Objective:To observe expressions of multidrug resistance-associated protein 1(MRP1)and glucose transporter-1(GLUT1)in osteosarcoma and their relationships with clinicopathological features.Methods:The clinical data of 60 patients with osteosarcoma from October 2015 to October 2016 were retrospectively analyzed.They were divided into study group(60 specimens of osteosarcoma tissue)and control group(60 specimens of normal bone tissue adjacent to the tumor,>5 cm from the edge of the tumor).All tissue samples were tested by immunohistochemistry.The expression levels of MRP1 and GLUT1 were detected.The expression levels of MRP1 and GLUT1 were compared between the two groups,and the relationships among the expression levels of MRP1 and GLUT1 and the clinicopathological features,and the prognostic survival rate of these patients were analyzed.Results:The positive expression rates of MRP1 and GLUT1 in the study group were significantly higher than those in the control group,and the differences were statistically significant(P<0.05).The positive expression rates of MRP1 and GLUT1 protein in the osteosarcoma patients with the clinical stages ofⅡb-Ⅲ,small tumor diameter,moderately and low differentiation,and survival time<1 year were higher than those of the patients with the clinical stages ofⅠ-Ⅱa,5 time of the tumor’s long diameter larger than the bone,highly differentiation,and survival time≥1 year,and the differences were statistically significant(P<0.05).Further,the median survival time of the patients with positive expressions of MRP1 and GLUT1 was significantly shorter than that of the patients with negative expressions,and the difference was statistically significant(P<0.05).Conclusions:MRP1 and GLUT1 are highly expressed in the osteosarcoma tissues and are related to the clinical stage,tumor size,degree of differentiation and survival time.It may play an important role in the occurrence and development of myeloma,or may provide a new target for the treatment of myeloma.
