摘  要:目的:通过观察大黄素对慢性压迫性损伤(chronic constriction injury,CCI)大鼠模型脊髓组织中髓鞘相关蛋白的影响,探讨其是否具有神经保护作用。方法:30只SD大鼠随机分为假手术组、CCI模型组、大黄素组3组,测量不同组别大鼠机械缩足反射阈值(mechanical withdrawal threshold,MWT)及热刺激缩足反射阈值(thermal withdrawal threshold,TWL),评估大黄素对CCI大鼠疼痛相关指标的改善情况,进一步采用RT-PCR及Western blot检测脊髓中髓鞘碱性蛋白(myelin basic protein,MBP)及髓鞘蛋白P0(myelin protein zero,MPZ)mRNA及蛋白表达水平。结果:术后11 d开始大黄素可改善CCI大鼠痛觉过敏,RT-PCR及Western blot结果显示,模型组大鼠脊髓中MBP、MPZ表达较假手术组显著降低,大黄素可升高MBP、MPZ的表达。结论:大黄素能上调MBP、MPZ髓鞘相关蛋白表达,从而调节神经损伤后髓鞘的形成,发挥改善神经痛的作用。Objective:To explore the effects of emodin on myelin proteins expression in chronic constriction injury(CCI)model.Methods:30 SD rats were randomly divided into sham group,CCI group and emodin group.Mechanical withdrawal threshold(MWT)and thermal withdrawal threshold(TWL)were measured to assess the improvement of pain symptoms on CCI model.The protein and mRNA expression of myelin basic protein(MBP)and myelin protein zero(MPZ)were measured by RT-PCR and western blot.Results:The behavior results showed that emodin can improve hyperpathia symptoms of CCI model on the 11th day after operation.The western blot and RT-PCR results showed that the expression of MBP and MPZ in CCI group were lower than that in sham group(P<0.05),and that the expression in emodin group were higher than that in CCI group(P<0.05)after treatment.Conclusion:The conclusion is that emodin can up-regulate the expression of MBP and MPZ to improve hyperpathia symptoms.
