摘 要:目的探讨石蒜碱对肝癌细胞增殖及自噬的影响,并分析其机制。方法采用CCK-8法分析石蒜碱对肝癌细胞增殖的抑制作用,MDC染色法检测石蒜碱对HepG-2细胞自噬的影响,Western blot检测石蒜碱对HepG-2细胞自噬相关蛋白Beclin-1、LC3 II、Mek/Erk信号通路关键因子Mek、Erk蛋白的影响,运用自噬抑制剂(3-MA)研究自噬对细胞增殖的影响。数据通过用SPSS 19.0进行统计分析。结果 CCK-8结果提示石蒜碱对肝癌HepG-2细胞的增长具有抑制作用(F=87.362,P<0.001);MDC染色法显示石蒜碱促进自噬小泡的形成。Western blot结果显示石蒜碱上调Beclin-1、LC3 II水平抑制自噬,下调Mek、Erk2磷酸化水平(p-Mek、p-Erk2)抑制HepG-2细胞增殖,差异均有统计学意义(F=25.571,P<0.001;F=315.002,P<0.001;F=35.481,P<0.001;F=150.103,P<0.001);与100μmol/L石蒜碱组相比,3-MA+石蒜碱组LC3II蛋白水平明显下调,p-Mek、p-Erk2蛋白水平明显上调(t=5.988,P=0.004;t=2.883,P=0.045;t=4.157,P=0.014)。结论一定浓度的石蒜碱能抑制肝癌HepG-2细胞增殖,可能与调控Mek/Erk信号通路诱发自噬有关。Objective To explore the effect of lycosine on the proliferation and autophagy of hepatic carcinoma and analyze its mechanism.Methods CCK-8 method was used to analyze the inhibitory effect of lycoline on the proliferation of hepatocellular carcinoma HepG-2 cells.Western blot analyzed the effects of lycologne on the autophagy-related proteins Beclin-1,LC3 II,and Mek/Erk signaling pathway p-Mek and p-Erk2 proteins in HepG-2 cells.Autophagy inhibitors(3-MA)were used to study the effects of autophagy on cell proliferation.The data was analyzed by SPSS 22.0 statistical software.Results CCK-8 results suggested that lycosine had an inhibitory effect on the growth of Hepatic carcinoma HepG-2 cells(F=87.362,P<0.001).MDC staining showed that lycosine promoted the formation of autophagosome.Western blot results showed that lycosine inhibited autophagy by up-regulating the levels of Beclin-1 and LC3 II,and inhibited the proliferation of HepG-2 cells by down-regulating the phosphorylation levels of Mek and Erk2(p-Mek,p-Erk2)(F=25.571,P<0.001;F=315.002,P<0.001;F=35.481,P<0.001;F=150.103,P<0.001).Compared with 100μmol/L lycoline group,the level of LC3II protein was significantly decreased,and the levels of p-Mek and p-Erk2 protein were significantly increased in the 3-MA+lycoline group(t=5.988,P=0.004;t=2.883,P=0.045;t=4.157,P=0.014).Conclusion A certain concentration of lycosine can inhibit the proliferation of HepG-2 cells,which may be related to the regulation of Mek/Erk signaling pathway-induced autophagy.
