摘 要:目的:探讨结肠癌组织中CD103和E-钙黏蛋白表达变化及其在结肠癌发展中的作用。方法:采用实时荧光定量PCR(qRT-PCR)和Western blot方法检测45例结肠癌、38例正常结肠组织中CD103和E-钙黏蛋白表达情况;体外实验中,敲低结肠癌细胞SW480 E-钙黏蛋白的表达,利用Transwell体外细胞侵袭实验观察SW480侵袭能力的变化;从结肠癌组织中分离肿瘤浸润淋巴细胞(TIL),并转染CD103过表达质粒,与SW480细胞混合培养,CCK8检测SW480细胞存活,评估TIL杀伤结肠癌细胞的能力。结果:与正常组织相比,E-钙黏蛋白在结肠癌组织中表达降低(P<0.001),CD103表达增加(P<0.001);E-钙黏蛋白表达与结肠癌有无淋巴结转移、临床分期以及分化程度有关,与无淋巴结转移患者相比,淋巴转移患者中E-钙黏蛋白表达更低;临床分期Ⅲ-Ⅳ期以及低分化患者中E-钙黏蛋白表达更低(P<0.001)。CD103表达与结肠癌的临床分期及分化程度有关,在Ⅲ-Ⅳ期以及低分化患者中CD103表达更低(P<0.001);与无淋巴结转移患者相比,淋巴结转移患者中CD103表达降低,但无统计学意义(P>0.05);敲低E-钙黏蛋白可促进SW480细胞穿过Transwell基质膜;TIL过表达CD103增加对SW480杀伤活性;敲低E-钙黏蛋白降低TIL和CD103+TIL细胞对SW480杀伤活性。结论:CD103和E-钙黏蛋白在结肠癌的发展及侵袭转移中发挥重要作用。Objective:To investigate the expression of CD103 and E-cadherin in colon cancer and determine the role in the development of colon cancer.Methods:Real-time quantitative PCR(qRT-PCR)and Western blot were used to detect the expression of CD103 and E-cadherin in 45 colon cancers and 38 normal colon tissues.In vitro,colon cancer cells SW480 was transfected with E-cadherin plasmid.Transwell invasion assay was used to observe the invasive ability of SW480.Tumor infiltrating lymphocytes(TIL)were isolated from colon cancer tissues,transfected with CD103 overexpression plasmid,mixed with SW480 cells.CCK8 was used to detect SW480 cell survival.The ability of TIL to kill colon cancer cells was calculated.Results:Compared with normal tissues,E-cadherin expression was decreased in colon cancer tissues(P<0.001),CD103 expression was increased(P<0.001).E-cadherin expression were associated with colon cancer with or without lymph node metastasis,clinical stages and degree of differentiation.The expression of E-cadherin was downregulated in patients with lymphatic metastasis compared with patients without lymph node metastasis.E-cadherin expression was lower in clinical stageⅢ-Ⅳand poorly differentiated patients(P<0.001).CD103 expression was associated with clinical stage and differentiation of colon cancer.CD103 expression was lower in stageⅢ-Ⅳand poorly differentiated patients(P<0.001).CD103 expression was decreased in patients with lymph node metastasis compared with patients without lymph node metastasis,but there was no statistical significance(P>0.05).Knockdown of E-cadherin promoted SW480 cells to cross the Transwell matrix membrane.TIL overexpressing CD103 increased SW480 killing activity;knockdown of E-cadherin reduced TIL and CD103+TIL cells SW480 killing activity.Conclusion:CD103 and E-cadherin play an important role in the development and invasion of colon cancer.
