摘 要:目的探讨腹腔内注射白介素-17A(IL-17A)抗体与IL-17F抗体对葡聚糖硫酸钠(DSS)诱导的溃疡性结肠炎(UC)小鼠的影响。方法 48只小鼠,随机分为正常对照组、模型组、抗IL-17A组、抗IL-17F组,每组12只。除正常对照组外,其余3组小鼠均饲以DSS溶液。抗IL-17A组和抗IL-17F组于实验第2、4、6天腹腔内分别注射IL-17A抗体和IL-17F抗体。观察4组小鼠体重、疾病活动度指数(DAI)评分、结肠长度及炎症病理学评分变化情况。检测血清IL-17A、IL-17F水平及结肠组织中IL-17A和IL-17F蛋白表达情况。结果与正常对照组比较,模型组、抗IL-17A组、抗IL-17F组小鼠体重下降,DAI评分增高,结肠变短,结肠炎症病理学评分增高,血清和结肠组织中IL-17A和IL-17F蛋白含量增高(P<0.05,P<0.01)。抗IL-17F组与模型组、抗IL-17A组比较,DAI评分更低、结肠更长、结肠炎症病理学评分更低(P<0.05,P<0.01)。抗IL-17F组血清IL-17A、IL-17F及结肠组织IL-17F水平均低于模型组(P<0.05)。结论对于DSS引发的UC,使用IL-17F抗体能够减轻肠道炎症程度。Objective To evaluate the effect of IL-17A antibody and IL-17F antibody by intraperitoneal injection on ulcerative colitis(UC)induced by dextran sodium sulfate(DSS)in mice.Methods Forty-eight mice were randomly divided into normal control group,model group,anti-IL-17A group and anti-IL-17F group,with 12 mice in each group.Except the normal control group,the other three groups were fed with DSS solution.The anti-IL-17A and anti-IL-17F groups were intraperitoneally injected with IL-17A antibody and IL-17F antibody respectively at 2,4 and 6 days after initiation of the experiment.The body weight,disease activity index(DAI)score,colon length and pathological score of colitis were observed.The serum levels of IL-17A and IL-17F and the expression of IL-17A and IL-17F in colon tissue were detected.Results Compared with the normal control group,mice in model group,anti-IL-17A group and anti-IL-17F group had weight loss,increased DAI score,shortened colon,increased pathological score of colitis,and increased contents of IL-17A and IL-17F protein in serum and colon tissue(P<0.05,P<0.01).Compared with the model group and anti-IL-17A group,anti-IL-17F group had lower DAI score,longer colon,and lower pathological score of colitis(P<0.05,P<0.01).The levels of serum anti-IL-17A,anti-IL-17F as well as IL-17F of colon tissue in the anti-IL-17F group were lower than those in the model group(P<0.05).Conclusion IL-17F antibody can effectively alleviate the inflammatory response of UC induced by DSS in mice.
