摘  要:目的探讨DNA甲基转移酶1(DNA methyltransferase 1,DNMT1)mRNA,miR-221及miR-152在镍化合物诱导细胞恶性转化过程中的表达变化。方法运用体外常规染毒培养方法,以2.00μg/cm2染毒培养人支气管上皮细胞(16HBE)至第36代,构建镍化合物转化的16HBE细胞;以软琼脂糖实验及裸鼠成瘤实验分析细胞转化情况;运用qRT-PCR方法分析染毒培养的第0代,20代和转化细胞(第36代)中DNMT1 mRNA,miR-221及miR-152的表达变化,并分析其作用及机制。结果在镍化合物染毒培养第36代的转化细胞中DNMT1 mRNA及miR-221表达水平明显高于第0代和20代细胞;而miR-152表达水平明显低于第0代和20代细胞。结论镍化合物诱导细胞恶性转化与DNMT1 mRNA、miR-221及miR-152的异常表达有关。Objective To investigate the alteration of expression levels of DNMT1 mRNA, miR-221 and miR-152 during the process of cell transformation induced by nickel compounds. Methods Nickel compound with the dose of 2.00 μg/cm^2 was used in human bronchial epithelial cell(16 HBE) transformation assay lasted for 36 passages. Soft agarose test and tumorigenesis experiment on nude mice were used to identify the transformation of 16 HBE cells. The expression levels of DNMT1 mRNA, miR-221 and miR-152 of the 0 th, 20 th and 36 th generations of cultured 16 HBE cells were analyzed by using qRT-PCR. Results The expressions level of DNMT1 mRNA and miR-221 of the 36 th generation of transformed cells were significantly higher than that of the 0 th and 20 th generations of cells, while the expression level of miR-152 was significantly lower than that of the 0 th and 20 th generations of cells. Conclusion The abnormal expressions of DNMT1 mRNA, miR-221 and miR-152 are related to the cell transformation induced by nickel compounds.
