摘 要:目的探讨α-突触核蛋白(α-Syn)过表达对帕金森病(PD)小鼠黑质多巴胺受体D1(D1DR)表达的影响。方法选取雄性C57BL/6J小鼠16只,随机将小鼠分为PD组和对照组,每组各8只。采用脑纹状体注射α-Syn预成型纤维(α-Syn PFF)法建立PD模型,对照组以相同方法注射PBS。各组小鼠于造模前1周及造模后4周,分别采用旷场实验、平衡杆实验、爬杆实验观察小鼠运动轨迹、运动速度及运动时间,采用Western blot方法检测小鼠黑质区D1DR、D2DR、α-Syn、酪氨酸羟化酶(TH)蛋白相对表达水平,采用免疫荧光法检测小鼠黑质区D1DR及TH表达变化。结果与对照组比较,PD组小鼠出现旷场实验运动速度减慢,平衡杆和爬杆耗时增加等运动迟缓行为学表现(P<0.05);Western blot结果显示,与对照组相比,PD组小鼠黑质α-Syn蛋白表达增加,D1DR及TH蛋白表达下降(P<0.05)。免疫荧光结果显示,PD组小鼠黑质TH和D1DR阳性神经元荧光强度均明显低于对照组。结论D1DR表达下降可能参与了α-Syn过表达所致的PD小鼠运动障碍。Objective To investigate the effect ofα-synuclein(α-Syn)overexpression on the expression of dopamine receptor D1(D1DR)in the substantia nigra of mice.Methods Sixteen male C57BL/6J mice were randomly divided into a Parkinson's disease(PD)group and a control group with eight mice in each group.The PD model was established by injectingα-Syn preformed fiber(α-Syn PFF)into the striatum,and PBS was injected into the control group in the same way.One week before and four weeks after modeling,open-field test,balance bar test and climbing-pole test were used to observe the movement track,movement speed and movement time of mice in each group.The relative expression level of D1DR,D2DR,α-Syn,tyrosine hydroxylase(TH)in the substantia nigra were detected by western blot,and the expression changes of D1DR and TH in the substantia nigra were detected by immunofluorescence.Results Compared with the control group,PD group mice showed slower movement speed in the open-field test,increased time consumption in balance bar and climbing-pole tests(P<0.05).Western blot results showed that compared with the control group,the expression ofα-Syn protein in the substantia nigra increased,and the expression of D1DR and TH protein decreased in the PD group(P<0.05).Immunofluorescence results showed that the fluorescence intensity of TH and D1DR positive neurons in the substantia nigra of the PD group was significantly lower than that of the control group.Conclusions The decreased expression of D1DR may be involved in the movement disturbance of PD mice induced byα-Syn overexpression.
