摘 要:通过香菇(Lentinula edodes)L808P65、EFISAAS5053P1和YAASM363P1原生质体单核菌丝体两两单单杂交,观察杂交过程中的菌落形态变化,利用ITS和rrnL部分序列分别鉴定双核菌丝体的细胞核和细胞质来源,以明确单单杂交过程中核迁移路径和细胞质遗传规律。结果表明:双亲菌落接触部位存在一条明显比周围菌丝体稀疏的分界线,3个亲本菌落的扩大速度从快到慢依次为EFISAAS5053P1、L808P65和YAASM363P1,双亲菌落接触区域的分界线均向菌落扩大速度慢的亲本菌落弯曲,有的甚至在分界线两端生长出两个与亲本菌落存在分界线的扇形菌落;细胞核迁移起始于分界线,双亲的细胞核互相向对方菌落迁移,不同培养皿中核迁移速度最快的区域不同;除位于分界线上的双核菌丝体外,左边亲本菌落的双核菌丝体细胞质来源于左边亲本菌落,右边亲本菌落的双核菌丝体细胞质来源于右边亲本菌落。Nuclear migration path and cytoplasmic inheritance during mon-mon crossing of Lentinula edodes were studied based on paired mon-mon crossings among three protoplast monokaryotic mycelia,L808 P65,EFISAAS5053 P1 and YAASM363 P1.Changes in colony morphology were observed and nuclear and cytoplasmic sources of dikaryotic mycelia were identified by ITS and partial rrnL sequences,respectively.The results showed that there was a clear demarcation line where two parent colonies contacted.Expansion speeds of three parent colonies from fast to slow were EFISAAS5053 P1,L808 P65 and YAASM363 P1 in order.Demarcation lines bent toward the parent colony with a relatively slower expanding speed and there were even two sectored colonies at both ends of a demarcation line.Nuclear migration began at a demarcation line with nuclei of two parent colonies migrated to each other.The fastest nuclear migration happened in different zones of different plates.Except dikaryotic mycelia developed on a demarcation line,dikaryotic mycelia on the left inherited cytoplasm from the parent on the left and dikaryotic mycelia on the right inherited cytoplasm from the parent on the right.
