摘  要:目的探讨消脂方对高脂饮食诱导肥胖小鼠糖代谢、胰岛素抵抗及慢性炎症的影响。方法将30只高脂饮食造模成功后的肥胖小鼠随机分为模型组、消脂方低剂量组、消脂方中剂量组、消脂方高剂量组、荷丹片组,每组6只,并选择C57BL/6J雄性正常小鼠6只作为正常组。模型组和正常组小鼠给予蒸馏水灌胃,消脂方低、中、高剂量组分别给予消脂方汤剂0.4583 g/mL、0.9165 g/mL、1.8330 g/mL灌胃,荷丹片组给予荷丹片药液0.0285 g/mL灌胃,均干预4周。观察比较各组干预后体质量、葡萄糖耐量实验血糖值、胰岛素抵抗指数(HOMA-IR)、葡糖糖曲线下面积(AUC)、胰岛素耐量实验血糖比值、肝组织中炎症因子[白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、白细胞介素-10(IL-10)]及相关蛋白(IKBα、p65、p-IKBα、p-p65)表达水平。结果消脂方各剂量组小鼠体质量、葡萄糖耐量实验各时间点血糖、AUC、FINS、HOMA-IR、胰岛素耐量实验血糖比值及IL-1β、IL-6、TNF-α水平均明显低于模型组(P均<0.05),30 min胰岛素/空腹胰岛素比值(0.5hINS/FINS)、60 min胰岛素/空腹胰岛素比值(1hINS/FINS)及血清IL-10水平均明显高于模型组(P均<0.05);除消脂方低剂量组p-p65蛋白及其磷酸化比值外,消脂方各剂量组p-IKBα、p-p65蛋白表达水平及两者磷酸化比值均明显低于模型组(P均<0.05)。消脂方中、高剂量组体质量和消脂方高剂量组葡萄糖耐量实验各时间点血糖值、AUC、FINS、HOMA-IR、胰岛素耐量实验血糖比值(除血糖90 min/血糖0 min)、IL-6均明显低于荷丹片组(P均<0.05),消脂方高剂量组0.5hINS/FINS、1hINS/FINS均明显高于荷丹片组(P均<0.05)。消脂方各剂量组之间以高剂量组各指标改善最明显,低剂量组最次,差异均有统计学意义(P均<0.05)。结论消脂方可能通过抑制NF-κB炎症信号通路,减轻炎症反应,从而改善高脂饮食诱导肥胖小�Objective It is to explore the effect of Xiaozhi Decoction on glucose metabolism,insulin resistance and chronic inflammation in obesity mice induced by high fat diet.Methods Thirty obese mice after successful high-fat diet modeling were randomly divided into model group,Xiaozhi Decoction low-dose group,Xiaozhi Decoction medium-dose group,Xiaozhi Decoction high-dose group,Hedan tablet group,6 mice in each group,and 6 C57BL/6J male normal mice were chosen as the normal group.The mice in the model group and the normal group were given distilled water by gavage,the ones in the Xiaozhi Decoction low,medium and high dose groups were respectively given Xiaozhi Decoction 0.4583 g/mL,0.9165 g/mL,1.8330 g/mL by gavage,and the onse in the Hedan tablets group were given Hedan tablets liquid 0.0285 g/mL by gavage,all the mice were intervened for 4 weeks.The body weight,blood glucose value in glucose tolerance test,insulin resistance index(HOMA-IR),area under the curve(AUC)of glucose,blood glucose ratio in insulin tolerance test,and the expression of inflammatory factors[interleukin-1β(IL-1β),interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),interleukin-10(IL-10)]and related proteins(IKBα,p65,p-IKBα,p-p65)in liver tissue in each group were observed and compared.Results The body mass,blood glucose of every time point in glucose tolerance test,AUC,FINS,HOMA-IR,blood glucose ratio in insulin tolerance test and the levels of IL-1β,IL-6,TNF-αof mice in each dose group of Xiaozhi Decoction were significantly lower than those in the model group(all P<0.05),30 min insulin/fasting insulin ratio(0.5hINS/FINS),60 min insulin/fasting insulin ratio(1hINS/FINS)and serum IL-10 levels were significantly higher than the model group(P<0.05).Except for the p-p65 protein and its phosphorylation ratio in the low-dose Xiaozhi Decoction group,the expression levels of p-IKBαand p-p65 protein and their phosphorylation ratio in each dose group of Xiaozhi Decoction groups were significantly lower than the model group(P<0.05).The body mass of t
