洛铂腹腔灌注化疗作用于进展期结直肠癌裸鼠动物模型的实验研究

摘　　要：目的:探讨洛铂腹腔灌注化疗对进展期结直肠癌裸鼠动物模型的治疗效果,从而为进展期结直肠癌的临床多途径治疗及预后等提供有效的实验依据。方法:将结直肠癌细胞株lovo种植于balb/c裸鼠腹腔中,将成瘤成功且符合实验标准的balb/c裸鼠再随机分为对照组(10只)、实验组(10只),对照组腹腔注射0.9%氯化钠溶液,实验组腹腔注射洛铂溶液(洛铂10mg/kg+5%GS)。两组每隔7d给药1次,每隔4d称量并记录裸鼠重量、观察并记录两组裸鼠存活情况,最后剥瘤称重并计算抑癌率。实验时间为50d。结果:经分析数据,对照组裸鼠肿瘤重量高于实验组(P<0.05),实验组抑癌率高于对照组(P<0.05),实验组裸鼠重量高于对照组(P<0.05);实验组平均生存时间高于对照组(P<0.05)。结论:洛铂腹腔灌注化疗对裸鼠进展期结直肠癌生长具有明显抑制作用并延长成瘤裸鼠的存活时间。Objective:To investigate the therapeutic effect of intraperitoneal infusion chemotherapy with carboplatin on advanced colorectal cancer in nude mice,so as to provide effective experimental basis for clinical treatment and prognosis of advanced colorectal cancer.Methods:The colorectal cancer cell line lovo was implanted into the abdominal cavity of balb/c nude mice,and the balb/c nude mice which were successful in tumor formation and in accordance with the experimental standard were randomly divided into control group(n=10)and experimental group(n=10).The control group was intraperitoneally injected with 0.9%sodium chloride solution and the experimental group was intraperitoneally injected with carboplatin solution(carboplatin 10mg/kg+5%GS).The two groups were given medicine every 7 days,weighed and recorded the weight of nude mice every 4 days,observed and recorded the survival of the two groups of nude mice,finally peeled off the tumor and weighed and calculated the tumor inhibition rate.The experimental time is 50 days.Results:According to the analytical data,the tumor weight of nude mice in the control group was higher than that in the experimental group(P<0.05),and the tumor inhibition rate in the experimental group was higher than that in the control group(P<0.05),the weight of nude mice in experimental group was higher than that in control group(P<0.05);the average survival time of experimental group was higher than that of control group.Conclusion:Cisplatin intraperitoneal infusion chemotherapy can significantly inhibit the growth of advanced colorectal cancer in nude mice and prolong the survival time of tumor-forming nude mice.
