摘  要:目的制备应用于表面增强拉曼光谱检测口腔鳞状细胞癌组织表皮生长因子(EGFR)表达的特异性纳米探针。方法通过化学还原法制备纳米Au颗粒,并在表面修饰高分子及EGFR抗体,制备特异性探针。检测特异性探针的光谱性能、细胞毒性,将其应用于小鼠鳞状细胞癌肿瘤组织内EGFR的表达检测,绘制肿瘤组织内EGFR拉曼光谱图像。结果通过化学还原法可以成功的将高分子巯基苯甲酸(MBA)、α-硫基-ω-羧基聚乙二醇(PEG)及EGFR抗体修饰于纳米Au粒子表面,构建纳米探针。该探针无细胞毒性,可以检测肿瘤组织内EGFR表达情况。结论MBA、PEG及EGFR抗体修饰的纳米探针可以应用于表面增强拉曼光谱检测口腔鳞状细胞癌肿瘤组织内EGFR表达。Objective To prepare a specific nanoprobe for detecting the expression of epidermal growth factor receptor(EGFR)for surface enhanced Raman spectroscopy of oral squamous cell carcinoma.Methods Au nanoparticles were prepared by chemical reduction method,and the surface was modified with macromolecule and EGFR antibody to prepare specific probes.The spectral performance and cytotoxicity of specific probes were detected,and they were applied to detect the expression of EGFR in mouse squamous cell carcinoma tumor tissues,and the Raman spectrum images of EGFR in tumor tissues were drawn.Results By chemical reduction method,macromolecular antibodies such as mercaptobenzoic acid(MBA),α-mercapto-ω-carboxyl polyethylene glycol(PEG)and EGFR could be successfully modified on the surface of Au nanoparticles to construct nanoprobes.There was no cytotoxicity in the probe,and EGFR expression in tumor tissues could be detected.Conclusions MBA,PEG and EGFR antibody modified nanoprobes can be used to detect EGFR expression in oral squamous cell carcinoma by surface enhanced Raman spectroscopy.
