摘 要:目的利用不同Rh表型的RhD阳性红细胞与系列倍比稀释抗-D试剂的反应结果,来评估不同Rh表型红细胞对抗-D效价测定的影响,为实验室检测抗-D效价的规范化操作提供依据。方法选取抗体筛查细胞和抗体鉴定细胞中不同Rh表型的RhD阳性红细胞,利用试管法和微柱凝胶卡法分别测定单克隆IgM抗-D试剂和IgG抗-D试剂的效价。结果R2R2、RZR2、R1~WR1谱细胞以及3个O型RhD阳性献血者混合红细胞测得的IgM抗-D和IgG抗-D效价最高,分别达到256和512,R0r和R1r测得的IgM抗-D和IgG抗-D的效价只有64和256。结论不同Rh表型的RhD阳性红细胞测得的抗-D效价有一定差异,各实验室应固定抗-D效价检测的指示细胞,建立本实验室的标准操作方法。Objective To evaluate the effects of red blood cells(RBCs)of different Rh phenotype on the determination of anti-D titer based on the findings of RhD(+)RBCs of different Rh phenotypes with serial dilution of anti-D reagent,so as to make the laboratory testing of anti-D titration more standardized.Methods RhD(+)RBCs of different Rh phenotypes were selected from the antibody screening cells and antibody identification cells,and the titers of IgM and IgG anti-D reagent were determined by tube and micro-gel card method,respectively.Results The titers of IgM and IgG anti-D measured by R2R2,RZR2,and R1WR1 spectrum cells and mixed RBCs of three RhD(+)donors with O-type were the highest,reaching 256 and 512,respectively.However,the titers of IgM and IgG anti-D measured by R0r and R1r were merely 64 and 256.Conclusion The anti-D titers yielded by RhD(+)RBCs of different Rh phenotypes were different.Laboratories should fix the indicator cells for anti-D titration and establish their own standard operating method.
