摘　　要：目的探讨miRNA-27a对人乳腺癌多柔比星耐药细胞株MCF-7/Adriamycin(MCF-7/ADR)的影响及其可能的作用机制。方法采用real-time PCR(RT-PCR)检测miRNA-27a和P-glycoprotein(P-gp)在乳腺癌细胞株MCF-7及MCF-7/ADR中mRNA表达。将miRNA-27a模拟物(mimics)转染到MCF-7/ADR细胞中,RT-PCR检测转染后miRNA-27a和P-gp的mRNA表达水平。Western blot法检测miRNA-27a模拟物转染后P-gp蛋白表达水平。细胞划痕实验检测转染0、24、48和72 h后细胞的迁移能力。结果miRNA-27a在MCF-7/ADR细胞中表达量低于MCF-7细胞,差异具有统计学意义(P<0.05)。上调miRNA-27a抑制MCF-7/ADR细胞中P-gp mRNA及蛋白的表达以及细胞的迁移能力,差异均具有统计学意义(均P<0.05)。结论miRNA-27a可通过抑制P-gp的表达水平逆转MCF-7/ADR细胞对多柔比星的耐药性。Objective To study the effect of miRNA-27a on adriamycin(ADR)-resistant human breast cancer cell line MCF-7/ADR and its possible mechanism.Methods The mRNA expression levels of miRNA-27a and P-gp in breast cancer cell lines MCF-7 and MCF-7/ADR were detected by real-time PCR.The miRNA-27a mimics were transfected into MCF-7/ADR cells,and the mRNA expression levels of miRNA-27a and P-gp were detected by real-time PCR.The expression level of P-gp protein after miRNA-27a mimics transfection was detected by Western blot.Cell scratch test was used to detect the migration ability of cells after transfection for 0,24,48 and 72 h.Results The expression level of miRNA-27a in MCF-7/ADR cells was lower than that in MCF-7 cells,and the difference was statistically significant(P<0.05).Up-regulation of miRNA-27a significantly inhibited the expression of P-gp mRNA and protein and the migration ability of MCF-7/ADR cells,with statistical significance(all P<0.05).Conclusion miRNA-27a can reverse the resistance of MCF-7/ADR cells to ADR by inhibiting the expression level of P-gp.
