摘 要:目的探讨姜黄素对急性坏死性胰腺炎(ANP)模型大鼠炎症反应及细胞自噬的影响。方法将18只雄性SD大鼠分为ANP组、姜黄素组和对照组,ANP组和姜黄素组使用注射牛黄胆酸钠的方法制造ANP模型,姜黄素组追加腹腔注射姜黄素,12 h后对大鼠胰腺组织进行病理学检查,并利用ELISA检测血清IL-6、TNF-α的含量,Western blot检测测胰腺LC3-Ⅱ的表达情况。结果病理组织学检查结果显示,对照组胰腺组织无明显病理变化,腺泡完整,叶间偶见水肿。ANP组和姜黄素组胰腺组织可见局部坏死,腺泡结构破坏,并可见红细胞及炎症细胞浸润。ELISA结果显示,IL-6、TNF-α在ANP组中相对于对照组上升,在姜黄素组中相对于ANP组中出现下降(P<0.05);ANP组及姜黄素组淀粉酶(AMY)明显高于对照组(P<0.05)。Western Blot结果显示姜黄素组、ANP组与对照组相比,LC3-Ⅱ的蛋白表达量均增加(P<0.05),且姜黄素组LC3-Ⅱ蛋白表达量显著高于ANP组(P<0.05)。结论姜黄素可减轻ANP模型动物的炎症反应并促进细胞自噬,可作为重症急性胰腺炎的治疗实验依据。Objective To investigate the effects of curcumin on autophagy and inflammatory response in rats with acute necrotizing pancreatitis(ANP).Methods The rats were divided into ANP group,curcumin group and control group.The rats in ANP group and curcumin group were intraperitoneally injected with curcumin.After 12 hours,pathological examination of pancreatic tissue was carried out.The levels of serum IL-6 and TNF-αwere detected by ELISA,and the expression of LC3-Ⅱin pancreas was detected by Western blot.Results Histopathological examination showed that there were no obvious pathological changes in the pancreas of the control group,with intact acini and occasional edema between the lobes.In ANP group and curcumin group,pancreatic necrosis,acinar structure destruction,and infiltration of red blood cells and inflammatory cells were observed.ELISA results showed that levels of IL-6 and TNF-αincreased in ANP group and were decreased in curcumin group(P<0.05).Western blot results showed that compared with the control group,the protein expression of LC3-Ⅱin curcumin group and ANP group was increased(P<0.05),and the protein expression of LC3-Ⅱin curcumin group was also higher than that in ANP group(P<0.05).Conclusion Curcumin can reduce the inflammatory reaction and promote autophagy in ANP model animals,which can be used as the experimental basis for the treatment of severe acute pancreatitis.
